摘  要:为了比较阿拉善双峰骆驼卵泡发育过程中颗粒细胞内蛋白质表达的变化,根据卵泡直径将颗粒细胞分为两类,利用双向电泳技术构建阿拉善双峰骆驼颗粒细胞蛋白质二维凝胶电泳图谱,凝胶经考马斯亮蓝染色后,以3倍以上的表达变化用PDQuest 8.0软件比较这两类颗粒细胞中蛋白质的表达差异。结果共检测到了12个差异点,经MALDI TOF/TOF-MS/MS鉴定发现它们是12种不同的蛋白质。其中丙酮酸脱氢酶E1、annexin A2可能与颗粒细胞能量代谢有关,这为阿拉善双峰骆驼颗粒细胞的研究奠定了理论依据。The aim of this study was to identify differentially expressed proteins in the granulosa cells of Alax Bactrian Camel during follicular development.Granulosa cells were divided into two groups according to their diameter.2-dimensional gel electrophoresis(2-DE) was performed to analyze the granulosa cell samples.Differential proteins were detected by PDQuest 8.0 software after stained with commassie blue.12 differentially expressed spots were observed.These spots were identified by MALDI TOF/TOF-MS/MS.12 unique proteins were be found.We hypothesize that pyruvate dehydrogenase E1 component subunit beta and annexin A2 play a role in the energy metabolism of granulosa cells.This study provides a solid basis for further study on granulosa cells of Alax Bactrian Camel.
