摘　　要：为了研究野菊花水煎剂对实验性免疫应激鸡血清中,促肾上腺皮质激素(ACTH)和皮质酮(CORT)含量及下丘脑中促肾上腺皮质激素释放激素(CRH)mRNA表达水平的影响。试验分为空白组、对照组和野菊花组,平均每组80只雏鸡。野菊花组于30日龄口服野菊花水煎剂,早晚两次,连续7 d。对照组和野菊花组32日龄胸肌肉注射接种10羽份的新城疫Ⅰ系疫苗,造成实验性免疫应激,空白组注射等量生理盐水。每组分别于免疫接种后1、3、5、7、14、21 d随机抓取10只鸡心脏采血并取下丘脑。双抗体一步夹心酶联免疫吸附法(ELISA)检测免疫应激后血清中ACTH和CORT的含量;实时荧光定量PCR法测定下丘脑CRH mRNA的表达水平。结果显示,野菊花能显著抑制血清中ACTH和CORT的含量及下丘脑中CRH mRNA的表达,对免疫应激有较好的缓解作用。The study was performed to investigate the effect of chrysanthemum indicum extract on levels of ACTH and CORT in serum and expression of hypothalamic CRH mRNA in immune stress broiler chicks.Test was divided into blank group,control group and wild chrysanthemum group.Each group contained 80 chickens.Chrysanthemum indicum group drank Chinese medicine decoction two times each day for 7 days.Control and Chrysanthemum indicum groups were vaccinated with 10 Newcastle disease vaccine by inteamuscular injection at age of 32-day-old,and blank group injected with saline.Eight chickens from each group were randomly selected to collect blood samples and took hypothalamus at day 1,3,5,7,14,and 21 post immunization.The content of ACTH and CORT were measured by the double antibody sand-wich enzyme-linked immunosorbent assay（ELISA）.The expression of CRH mRNA was measured by real-time quantitative PCR.The results showed that the content of ACTH and CGRT in serum and the expression of CRH mRNA after immunological stress were significantly increased.And Chrysanthemum indicum significantly inhibited ACTH and CORT in serum and the expression of CRH mRNA k in hypothalamus.
