摘 要:目的:建立豨莶草药材HPLC指纹图谱,并同时测定豨莶草中奇壬醇、豨莶苷及豨莶精醇3种主要活性成分的含量。方法:采用Kromasil WQC_8柱(4.6mm×250mm,5μm)色谱柱,以甲醇-乙腈-0.05mol/L KH_2PO_4溶液为流动相进行梯度洗脱,流速1.0 mL/min,柱温30℃,进样量10μL,检测波长215nm,记录不同产地10批豨莶草色谱图数据,并同时测定了其中3种主要活性成分的含量。结果:建立了豨莶草HPLC指纹图谱,10批不同来源的豨莶草药材指纹图谱相似度较高,并利用"中药色谱指纹图谱相似度评价软件"生成了豨莶草药材的对照指纹图谱,得到14个共有峰,各色谱峰分离度良好,符合指纹图谱检测要求。并成功测定了不同批次豨莶草中奇壬醇、豨莶苷及豨莶精醇3种主要活性成分的含量,结果分别为0.12%~1.41%、0.10%~2.19%、0.01%~0.39%。结论:本方法操作简便、准确、重现性好,为豨莶草药材的鉴别和质量控制提供科学依据,并为豨莶草活性成分的含量测定提供参考方法。Objective :To establish Siegesbeckia herb material HPLC fingerprint, and simultaneous determina- tion of Herba Siegesbeckiae kirenol, Siegesbeckia glycosides and Siegesbeckia alcohol 3 kinds of main active ingredient content. Methods:Kromasil WQ C8 column (4.6 mm× 250 mm, 5 m) column, methanol - acetonitrile (0.05 mol / L. KH2PO4 solution for flow by gradient elution, flow rate of 1.0 mL / min, column temperature of 30 DEG C, into the sample weight of 10 g 1, detection wavelength of 215 nm recorded from different regions of 10 batches of Herba Sieges beckiae chromatogram data, and simultaneous determination of the content of three kinds of active ingredients. Results: Herba Siegesbeckiae HPLC fingerprint were established. Ten batches of different sources of Siegesbeckia herb fingerprint similarity higher, and the traditional Chinese medicine chromatographic fingerprint similarity evaluation software to gen erate the control fingerprint of Herba Siegesbeckiae herbs, get 14 common peaks, all kinds of spectral peaks were well separated, in line with the fingerprint. And the successful determination of the content of different batches of Herba Sieg esheckiae kirenol, Siegesheekia glycosides and Siegesbeckia alcohol 3 kinds of main active components, the results were 0. 12% to 1.41%, 0.10% ~ 2.19%, 0.01% ~ 0.39%. Conclusion :This method operation is simple, accurate and repro- ducible, Siegesbeckia herb identification and quality control to provide a scientific basis, and Herba Siegesbeckiae active ingredient content determination to provide reference method.
