摘  要:目的探讨"GA118-16A型"国产法医遗传分析仪受环境温度变化影响的情况。方法以分子量内标Gene Scan^TM-500LIZ为对象,通过分型情况、迁移速度和重现性,检测不同温度环境对该国产法医遗传分析仪运行的影响。数据经统计分析,评判仪器适温性能。结果实验表明,1随着环境温度的提高,内标片段中250bp的实际读值逐渐靠近设定值,迁移速度加快;213个实验室温度下,标准品均可获得完整、清晰、准确的分型图谱,RSD在0.1%左右,数据离散程度小,在20-26℃间仪器对DNA分离重现性最好;3能承受2-5℃的温度波动,在23-27℃检测稳定性最佳。结论该仪器能满足环境温度范围18-30℃间的检测需求,重现性好,片段分型准确,运行稳定。Objective To explore genetic analyzer. Methods Using the the effect of ambient temperature GeneScan-500 LIZ internal size on the performance of GA118-16A standard as research object, we detected the influence of different environmental temperatures on the performance of domestic forensic genetic analyzer GA118-16A by observing typing, migration and reproducibility. Statistical analysis of the original data was Carried out in order to judge the temperature adaption performance of the instrument. Results The laboratory tests showed that: (1) with the increase of ambient temperature, actual value of fragment 250bp gradually closed to the setting value accompanied by the higher migration speed; (2) Complete, clear and accurate genotype profiles from standard samples were obtained under 13 different laboratory temperatures. When the ambient temperature was between 20℃ and 26℃, approximately 0.1% RSD, small data discrete degree and the best DNA separation repeatability were observed; (3) The instrument could withstand the temperature fluctuations of 2 to 5~C, and the best testing stability was observed when the ambient temperature was between 23℃ and 27℃. Conclusion The instrument can satisfy the test requirement in the range of temperature from 18 to 30℃, and is of good accuracy and reproducibility.
