摘 要:目的通过观察灵芝孢子粉对百草枯中毒大鼠肺组织及血清氧化应激和炎症因子水的影响,探索其对肺损伤的保护作用。方法将72只(SD)大鼠随机分为正常对照组(A组)、百草枯模型组(B组)、灵芝孢子粉治疗组(C组),B组和C组再分为1 h、6 h、24 h、72 h 4个亚组,每个亚组8只。A组给予生理盐水20 mg/kg一次性灌胃,B组和C组采用20%百草枯溶液20 mg/kg一次性灌胃,然后分别给予等容积的生理盐水、灵芝孢子粉150 mg/kg/q12 h灌胃。按时间点取股动脉血清测定IL-6水平,处死后取肺组织,观察病理变化,采用ELISA法测定肺组织中TGF-β1和TNF-a含量;应用黄嘌呤氧化酶法测定血清SOD活力。结果 B组、C组大鼠肺组织均出现大片出血、坏死,但C组大鼠肺组织损伤程度较B组显著减轻。与A组比较,B、C组肺组织1 h、6 h、24 h、72 h TGF-β1表达显著增加(P<0 01),但C组较B组为低(P<0.05)。与A组比较,B组及C组肺组织中TNF-a、血浆IL-6从第6 h开始显著增加(均P<O.01),但C组在各时点均较B组为低(均P<O.05)。C组在各时点的SOD活力均较B组为高(均P<0.05)。结论灵芝孢子粉能减轻百草枯对大鼠肺组织的损伤,其机制可能与抑制TGF-β1的表达、降低肺组织TNF-a含量及血清l L-6水平、提高SOD的含量有关。Objective: To investigate the effect of lingzhi spore on paraquat poisoned rats lung tissue and its mechanism. Methods: Seventy-two Sprague Dawley( SD) rats were randomly divided into normal control group( n = 8),paraquat model group( n = 32) and lingzhi spore treatment group( n = 32) Rats in model and treatment groups were administered with a single dose of PQ( 20 mg / kg) by gavage,then 150 mg / kg lingzhi spore( treatment group) or 0. 9% saline( model group)was administered by gavage. The rats in model and treatment groups were sacrificed at 1,6,24 and 72 h,after treatment,the blood samples were taken from femore arterial and the lung tissue samples were also collected. The serum IL-6 levels were measured. and the pathological changes in lung tissue were examined: the expression of TGF-β1、TNF-a were detected by enzyme linked immune-osorbent assay,the SOD levels in lung tissue were also measured by xanthine oxidase method. Results: Severe hemorrhage and necrosis were noticed pathologically in lung tissue of mode group,the degree of lung injury in treated group was attenuated compared with model group. The expression on TGF-β1 in lung tissue of both model and treatment groups at1,6,24,72 h increased significantly than the control group,but the expression in treatment group was lower than that in model group( P〈0. 05) The TNF-a levels and IL-6 levels of model and treatment groups began to increase from 6h after poisoninq,but those in treatment group were lower at each time points( P〈0. 05). SOD significantly decreased,but SOD in every point of treatment group were upon maturity of lingzhi spore only for high in the paraquat model group( P〈0. 05). Conclusion: lingzhi spore can attenuate the lung tissue injury induced by paraquat in rats,which is associated with down-regulation of TGF-β1,TNF-a expression in lung tissue and decreased serum IL-6 levels,improve the content of SOD and there by reduce paraquat injury of lung tissue of rats.
