摘 要:目的探讨复序橐吾和狭苞橐吾乙醇提取物体外抗肿瘤作用,为抗肿瘤药物研发以及多途径开发、利用长白山橐吾属植物资源提供参考依据.方法采用MTT法检测复序橐吾和狭苞橐吾乙醇提取物(FXEt OH、XBEt OH)在体外对2种肿瘤细胞(Hela,A549)增殖的抑制作用;应用流式细胞术检测细胞凋亡;以Vc做对照,采用邻二氮菲法、邻苯三酚自氧化法测定2种橐吾乙醇提取物对羟自由基()、超氧阴离子自由基(O-2·)的清除率.结果用不同质量浓度的2种橐吾乙醇提取物处理A549,Hela细胞48 h后,对Hela,A549细胞的抑制率均随样品浓度增加而增大;对的清除,FXEt OH,XBEt OH,Vc的IC50值分别为73.34,335.30,79.31μg/m L;对O-2·的清除,FXEt OH,XBEt OH,Vc IC50值分别为64.81,131.89,191.62μg/m L.结论复序橐吾和狭苞橐吾乙醇提物在体外均具有一定的抗肿瘤活性和较好的综合抗氧化能力,抗氧化可能是其抗肿瘤作用的机制之一.Objective To discuss the antitumor effects of ethanol extracts from Ligularia jaluensis and Ligularia intermedia in vitro,and to provide references for the development of antitumor drugs as well as the comprehensive utilization and development of Ligularia genus resources in Changbai Mountain. Method The inhibiting effects of ethanol extracts from Ligularia jaluensis and Ligularia intermedia on the multiplications of 2 kinds of tumour cells (Hela,A549) were determined by MTT colorimetric assay in vitro. Flow cytometry was applied to test cell apoptosis. The effects of ethanol extracts from two kinds of ligularia genus plants on the clearance of Hydroxy free radical( ) and superoxide anion free radical ( O-2 · ) were measured by Phenanthroline and Pyrogallol autoxidation methods. Results The A549 and Hela cells were treated with different concentrations of the ethanol extracts for 48 hours, and the inhibition rates increased remarkably with increasing of concentration. For the clearance of ,the IC50 of FXEtOH,XBEtOH and Vc were 73. 34,335. 30 and 79. 31μg/mL,respectively. For the clearance of O-2 ·, the IC50 of FXEtOH, XBEtOH and Vc were 64. 81, 131. 89 and 191. 62 μg/mL, respectively. Conclusion Both the ethanol extracts of Ligularia jaluensis and Ligularia intermedia possess the antineoplastic activity in vitro and good comprehensive antioxidant capacity which may be one of their possible mechanism on antitumor.
