脱酚棉籽蛋白培养腐熟用枯草芽孢杆菌DK36的研究

摘  要:以脱酚棉籽蛋白为氮源,研究了腐熟用枯草芽孢杆菌DK36的生长及产芽孢条件。结果发现,当脱酚棉籽蛋白为氮源时,菌体生长良好,可完全代替豆饼粉,甚至代替酵母粉。当葡萄糖为碳源时,枯草芽孢杆菌DK36不形成芽孢。当玉米粉作碳源时,枯草芽孢杆菌DK36可大量形成芽孢,其芽孢数和芽孢率分别达到2.7×109CFU/m L和93.4%。培养基中添加钙、镁、锰和钾盐时芽孢数和芽孢率均明显提高,而且钾盐与钙、镁和锰盐在促进生长和产芽孢上具有协同作用,其中协同效果最好的组合是KH2PO4与Mg SO4,其芽孢数和芽孢率分别为3.2×109CFU/m L和98.4%。然而,脱酚棉籽蛋白会造成芽孢形成缓慢,生产效率降低。In this work,cell growth and sporulation of Bacillus subtilis DK36 for microorganism composting preparations were investigated using degossypolized cottonseed protein as nitrogen source. The results showed that the cells grew very well when degossypolized cottonseed protein was used as nitrogen source,and it could replace soybean meal and even yeast extract. The cells of B. subtilis DK36 did not sporulate when glucose was used as carbon source. The cells produced a great amount of spores when corn meal was used as carbon source. The treatment with corn meal had the highest spore yield and sporulation efficiency,which were 2. 7 × 10^9 CFU / m L and 93. 4%,respectively. Supplement of Ca^2+,Mg^2+,Mn^2+and K^+ improved significantly spore yield and sporulation of B. subtilis DK36. K^+ had a synergism with Ca^2+,Mg^2+and Mn^2+in stimulation of the cell growth and sporulation. KH2PO4 and Mg SO4 had the best synergism effect,and its spore yield and sporulation efficiency were 3. 2 × 10^9 CFU / m L and 98. 4% respectively. However,degossypolized cottonseed protein made sporulation of the cells very slowly,which greatly reduced the production efficiency.
