地丁豆根白术汤抗肿瘤作用的实验研究

摘　　要：目的研究地丁豆根白术汤的抗肿瘤作用。方法用地丁豆根白术汤复方中药水提取物作用肿瘤细胞(sp2/0)和正常细胞(CEF)24 h,显微计数研究复方中药对肿瘤细胞的杀伤作用;流式细胞技术和AnnexinⅤ-FITC/PI探讨复方中药对肿瘤细胞的杀伤机制;腹腔注射sp2/0细胞建立小鼠同源腹水型肿瘤模型,灌胃给药,研究复方中药对发病小鼠的治疗作用。结果 2.4 mg/m L的复方中药水提取物作用24 h后,可完全杀灭sp2/0细胞,而对CEF细胞无明显杀伤作用;2.4 mg/m L的复方中药可通过诱导细胞凋亡的方式杀伤肿瘤细胞,药物作用2 h时凋亡比率达到峰值(58.77%);60 mg/d的药量能明显减少小鼠腹腔肿瘤的数量、大小,明显延长小鼠寿命。结论该复方中药对恶性肿瘤有一定的控制和治疗作用,可以作为治疗恶性肿瘤的药物做进一步研究。Objective To research the effect of Chinese herbal complex（CHM） on fight against the cancer. Methods In this study, we investigated the effects of CHM to tumor cell on normal cell lines CEF and cancer cell lines sp2/0 for 24 h.We used flow cytometry and AnnexinⅤ-FITC/PI to study the effects of CHM on apoptosis of sp2/0 cells. Mouse tumor homograft model were constructed and treated by feeding CHM water extract. Results sp2/0 exposure to 2.4mg/m L of CHM water extract could suppress the proliferation after 24 h, and CEF was growing well. The CHM could significantly induce sp2/0 cell apoptosis and the apoptotic rate was 58.77% after 2 h. The experiments in vivo showed that the dosage of CHM（60mg/d） could inhibit growth of tumor obviously and prolong life of tumor-bearing mice. Conclusion This CHM has some effects in control and treatment in malignant tumor, and might be worthy of further study for its potential as a therapeutic agent for malignant tumor treatment.
