摘 要:甘草多糖是甘草提取物中的重要活性组分,具有免疫调节和抗氧化等多种药理作用,对其进行分离纯化的研究具有重要意义。研究探索了一种利用晶胶介质分离甘草多糖的新途径。以预处理和水提甘草根粉末得到的粗多糖为原料,用甲基丙烯酸二甲氨乙酯接枝聚丙烯酰胺晶胶基质制备得到的阴离子交换晶胶介质,对甘草多糖进行了快速层析分离,得到了精制的甘草多糖,确定了适宜的工艺条件。结果表明,阴离子晶胶介质可以吸附甘草多糖,其层析分离迅速;以去离子水为床柱平衡、冲洗及洗脱液配置的基础液,在流速1 cm·min-1条件下,晶胶介质对甘草多糖的吸附容量达670μg·m L-1晶胶,说明晶胶层析可作为天然药用多糖快速分离的一种新方法。Glycyrrhiza polysaccharides are important bioactive compounds in Glycyrrhiza roots with immunomodulation and potential therapeutic activities. It is of great importance to investigate related separation and purification methods. A novel ion-exchange chromatographic method using monolithic cryogels for rapid isolation of Glycyrrhiza polysaccharides from aqueous extract feedstock of Glycyrrhiza roots was developed. The crude Glycyrrhiza polysaccharides were extracted from pre-treated Glycyrrhiza roots, and then polyacrylamide-based anion-exchange monolithic cryogels grafted with N,N'-dimethylaminoethyl methacrylate was employed to purify the polysaccharides. The chromatographic conditions were optimized and the results show that the polysaccharides are effectively adsorbed by the cryogels. Using deionized water as the equilibrium, wash and running liquids and 1.0 mol·L^-1 Na Cl as the elution liquid, the maximum adsorption capacity of Glycyrrhiza polysaccharides on the cryogels is 670 μg·m L^-1 under a flow rate of 1 cm·min^-1, which indicates that the present method could be a potential approach for the rapid separation of polysaccharides from aqueous herbal feedstock.
