摘 要:目的评估近红外激光及PEG-IR-780-C13热消融治疗肾癌的效果。方法建立小鼠原位肾癌模型,通过尾静脉将PEGIR-780-C13注射到小鼠体内,利用小动物成像系统观察PEG-IR-780-C13在小鼠主要器官中的分布。近红外激光照射小鼠肾癌部位,利用可视近红外稳定测定仪测定小鼠肾癌部位温度变化。通过小鼠肾癌体积变化及HE染色评估近红外激光及PEG-IR-780-C13热消融肾癌的效果。结果 PEG-IR-780-C13注射后能够在肿瘤部位蓄积,近红外激光照射后肿瘤部位稳定明显上升。近红外激光及PEG-IR-780-C13能够导致肿瘤组织坏死,治疗组肿瘤体积明显减小。结论近红外激光及PEG-IR-780-C13能够高效的产生热能杀伤肿瘤,为热消融肾癌提供了新的探索方向。Objective To evaluate the efficacy of PEG-IR-780-C13 and near-infrared(NIR)irradiation for thermal ablation of renal carcinoma by using an in situ tumor model.Methods Mouse models of in situ carcinoma were established.PEG-IR-780-C13 was injected through tail vein.The distribution of PEG-IR-780-C13 in the major organs of the mice were observed with IVIS Lumina imaging system.The changes of temperature in the tumor tissues during NIR irradiation were measured using Visual IR thermometer.The effect of NIR laser and PEG-IR-780-C13 on renal tumors was evaluated by measuring the size of tumor and HE staining.Results PEG-IR-780-C13 accumulated in renal carcinoma after systematic delivery.Upon NIR irradiation,the temperature of renal carcinoma tissues increased significantly.NIR irradiation and PEG-IR-780-C13 induced necrosis and decreased the size of renal carcinoma.Conclusion NIR irradiation and PEG-IR-780-C13 could generate tumor-specific heat for the destruction of renal carcinoma,providing a novel strategy for thermal ablation.
