摘 要:目的:用Meta分析的方法综合评价Toll样受体4(TLR4)基因Asp299 Gly多态性与2型糖尿病(T2MD)的关系,为探索T2MD的遗传背景提供循证医学证据。方法:系统检索Pub Med,万方数据库、维普信息资源系统、中国期刊全文数据库及中国生物医学文献数据库,全面收集2015年7月之前发表的TLR4基因多态性与T2MD相关性的病例对照研究。对资料进行异质性检验,以确定采用固定模型或随机模型进行合并分析。结果:本研究共纳入5篇文献,累计T2MD 2 199例,对照组1414例。Meta分析结果显示,TLR4基因Asp299Gly位点的基因型Meta分析结果合并OR值为1.05,95%CI为(0.85-1.29),P=0.68差异无统计学意义;TLR4基因Asp299 Gly位点的等位基因Meta分析结果合并OR值为1.05,95%CI为(0.86-1.28),P=0.66,差异无统计学意义。结论:TLR4基因Asp299Gly多态性与T2MD易感性无关联。AIM: To explore the association be- tween TLR4 gene Asp299Gly polymorphism and the susceptibility to type 2 diabetes mellitus ( T2MD ), and to provide basis for evidence-based medicine for the genetic background of T2MD. METHODS: Electronic search strategy was carried out, using Pub Med, Wan fang Database, China National Knowledge Infrastructure (CNKI) and CBM from inception to July 2015 to collect data on the association between TLR4 gene Asp299Gly polymorphism and the susceptibility to T2MD. Fixed effects model or random effects model was employed according to statistical tests for homogeneity. Publication bias was assessed by rank correlation test. RESULTS:A total of 5 studies were available for this analysis, including 2 199 cases and 1 414 controls. The metaanalysis results of Asp299Gly genotype showed OR = 1.05, 95% CI = 0.85 - 1.29, P = 0.68 and the allele results were OR = 1.05, 95% CI = 0.86 -1.28, P=0.66. CONCLUSION : TLR4 Asp299Gly polymorphism may be not associated with T2MD.
