摘  要:目的研究丝状真菌SIIA-F1108发酵液中抗真菌的活性成分。方法利用大孔吸附树脂吸附、正相硅胶色谱层析和高压液相制备等技术,结合白念珠菌抑菌实验进行跟踪检测,分离丝状真菌SIIA-F1108的发酵液中的活性组分;通过紫外光谱、红外光谱、高分辨质谱、核磁共振进行分析,确定活性组分的结构。结果分离纯化得到SIIA-C1108A和SIIA-C1108B活性组分,分子式均为C_(50)H_(80)N_8O_(17),与文献报道的纽莫康定(pneumocandin)B_0和C_0同质。结论从丝状真菌SIIA-F1108的发酵液中分离得到的两个具有抗真菌活性物质分别为纽莫康定B_0和C_0,其中纽莫康定B_0作为合成醋酸卡泊芬净的前体物质,具备重要的经济价值。Objective To study the active compounds which inhibit the growth of fungi from the fermentation broth of filamentous fungi SIIA-F1108. Methods The active components were isolated from the fermentation broth of filamentous fungi SIIA-F1108, using the technologies such as normal phase silica gel chromatography and preparative HPLC, combed with the fungistatic test against Candida albicans for detection. The structure characteristics of these active compounds were finally identified through the elemental analysis and spectral analysis, including ultraviolet spectrum, infrared spectroscopy, high resolution mass spectrum and nuclear magnetic resonance spectroscopy. Results The active compounds SIIA-C1108A and SIIA-C1108B were separated, which molecular formula were both the C50H80N8O17. They were confirmed to be homogeneous with pneumocandin B0 and C0. Conclusion The two antifungal compounds isolated from the fermentation broth of filamentous fungi SIIA-F 1108 were confirmed to be pneumocandin B0 and Co. Furthermore, pneumocandin Bo is the precursor of caspofungin acetate which has the enormous economic value.
