摘 要:目的:从Wnt/β-catenin信号通路观察透骨消痛胶囊对大鼠膝骨关节炎关节软骨的保护作用。方法:60只健康清洁级2月龄雄性SD大鼠,随机分为正常对照组、模型对照组和透骨消痛胶囊组,每组20只。模型对照组和透骨消痛胶囊组采用质4%木瓜蛋白酶双膝关节腔注射,成功复制骨关节炎模型后,透骨消痛胶囊组给予透骨消痛胶囊287.5 mg·kg^(-1),正常对照组和模型对照组给予相同剂量的生理盐水。给药2个月后,采用Western blot法观察各组关节软骨中含Ⅰ型血小板结合蛋白基序的解聚蛋白样金属蛋白酶-5(ADAMTS-5)、Wnt-4、β-catenin和基质金属蛋白酶-13(MMP-13)的表达。结果:与正常对照组比较,模型对照组ADAMTS-5、Wnt 4、β-catenin和MMP-13表达升高(P<0.05);与模型对照组比较,透骨消痛胶囊组ADAMTS-5、Wnt 4、β-catenin和MMP-13表达降低(P<0.05)。结论:透骨消痛胶囊对关节软骨的保护作用与其降低Wnt/β-catenin信号通路表达相关。Objective:To investigate the protective effect of Tougu Xiaotong Jiaonang(透骨消痛胶囊)on knee articular cartilage of rats from Wnt/β-catenin signaling pathway.Methods:Sixty healthy twomonth male SD rats were randomly divided into a normal control group,a model control group and a Tougu Xiaotong Jiaonang group(TXJ group),20 rats in each.The model control group and the TXJ group were given double knee joint cavity injection with 4% papain.After the osteoarthritis models were successfully established,the TXJ group was given Tougu Xiaotong Jiaonang 287.5 mg·kg-1,while the normal control group and the model control group were given the same dose of physiological saline.After 2 months of administration,the Western blot method was used to observe the expressions of ADAMTS-5,Wnt-4,β-catenin and MMP-13 in articular cartilage of three groups.Results:Compared with the normal control group,the expressions of ADAMTS-5,Wnt-4,β-catenin and MMP-13 in the model control group increased(P 〈 0.05),while compared with the model group,the expressions of ADAMTS-5,Wnt-4,β-catenin and MMP-13 in the TXJ group decreased(P 〈 0.05).Conclusion:The protective effect of Tougu Xiaotong Jiaonang on articular cartilage is correlated with the reduction of the Wnt/β-catenin signaling pathway expression.
