摘 要:目的探讨ABO血型鉴定中正反定型不一致时,采用基因测序方法鉴定血型的意义。方法对本血液中心在ABO血型鉴定时发现的无偿献血者正反定型结果不一致样本,采用试管法进行血型血清学分析和亚型鉴定;对血清学检测为ABO亚型的样本,再采用PCR-SBT方法对6、7外显子和部分内含子进行基因序列分析。结果 2例血清学鉴定为A亚型B的样本,经ABO基因测序确定基因型为Cis AB/B,分别为Cis AB01/B101和Cis AB05/B101。结论由于B基因的作用,Cis AB血型经血清学方法难以被发现并确定,基因测序有助于更准确地鉴定血型。Objective To identify the true ABO subgroup with gene sequencing when sera and cell experiments show disagreement results. Methods We screened ABO blood type from volunteers of blood donation in the blood center. The discrepant samples in experiments need serological analysis to verify ABO subgroup. Mutations of complete 6 and 7 exons including parts of introns were screened by direct sequencing. Results Two A subgroup / B samples in serological test are determined as Cis AB01 / B101 and Cis AB05 / B101 subgroup,respectively. Conclusion It is difficult to identify Cis AB when B gene plays a part in phenotyping and DNA sequencing rather than serological testing is able to clarify the phenotype.
