太子参同源四倍体表型变异的ISSR分析

摘  要:利用前期研究建立的太子参ISSR-PCR体系,从100条ISSR引物中筛选出15条适于太子参扩增的引物.对2份二倍体太子参原种及经染色体加倍形成的19个同源四倍体太子参表型变异株系进行ISSR分子标记分析,将二倍体与其同源四倍体株系以及同源四倍体株系之间的差异进行对比分析.结果发现只有个别四倍体株系相对于二倍体原种表现出少量的基因差异,且同源四倍体株系间极少表现出基因差异.表明在太子参染色体加倍的过程中发生了低频率的基因突变;但是鉴于同源四倍体株系的性状变异较大,推测这些性状变异并不都是基因突变造成的;基因突变可能不是表型变异的主要因素,其他因素对表型变异的贡献可能更大.Based on previous studies of Pseudostellaria heterophylla( Miq.) Pax ISSR-PCR system, 15 ISSR primers which were suitable for Pseudostellaria heterophylla( Miq.)Pax were screened from the 100 ISSR primers. Two diploid original species and 19 autotetraploid phenotypic variation strains which were produced by chromosome reduplication were analysed by ISSR molecular marker technique. The differences between diploid original species and autotetraploid strains and autotetraploid strains each other were analysed. The results showed that only a few autotetraploid strains showed small genetic differences from diploid original species.Autotetraploid strains rarely showed genetic differences with each other, which was also found at the same time. These results indicated that low frequency gene mutations took place during chromosome reduplication. Given that autotetraploid strains showed significant phenotypic variation, it suggested that these phenotypic variation were not caused by gene mutation completely. Gene mutation may not be the main factor of phenotypic variation. The contribution of other factors on the phenotypic variation may be more important.
