摘 要:为了观察17-β-雌二醇对猪卵母细胞体外成熟及体外受精胚的影响,选用屠体卵巢上的卵母细胞为试验材料,将其分为4组,分别在培养基中添加0、25、50、75μmol/L 17-β-雌二醇,体外成熟培养时间为36~44 h,受精胚培养时间为9 d。结果表明,在培养基中分别添加17-β-雌二醇0、25、50、75μmol/L时猪卵母细胞体外成熟率分别为58.5%、67.1%、80.9%、64.5%,卵裂率分别为51.2%、56.5%、63.5%、59.2%,囊胚形成率分别为16.7%、24.2%、19.5%、24.0%。结果显示在培养基中添加50μmol/L 17-β-雌二醇可提高猪卵母细胞体外成熟率至80.9%,提高体外受精胚的卵裂率至63.5%,但囊胚形成率以添加量为25μmol/L的组最高,说明添加17-β-雌二醇可提高猪卵母细胞成熟率、卵裂率以及囊胚率。For observing the effect of 17-β-E2 on the porcine oocytes maturation and fertilized embryos' development in vitro, the porcine oocytes collected from prepubertal gilts' ovaries were cultured in maturation medium which added different concentration 17-β-estrodiol (0,25,50.75 μmol/L) for 36-44 hours. The fertilized eggs were cultured for 9 days. The results showed that the maturation rate of each addition was 58.5%, 67.1%, 80.9%, 64.5%, respectively. The cleavage rate was 51.2%, 56.5%, 63.5%, 59.2%, respectively, and the blastocyst rate was 16.7%, 24.2%, 19.5% and 24.0%, respectively. It indicated that the addition of 17-β-estrodiol could improve maturation rate, cleavage rate and blastocyst rate, and the maturation rate & cleavage rate of 50 μmol/L addition were higher than the other 3 groups, but the blastocyst rate of 25 μmol/L addition was higher than the other groups.
