摘 要:目的探讨麻黄细辛附子汤(MXF)对正常小鼠外感和肾阳虚模型小鼠外感干预作用效果差异。方法KM小鼠按体质量和肛温随机分成6组,并设空白对照组。腹腔注射苯甲酸雌二醇(8 mg/kg)法复制肾阳虚小鼠模型,流感病毒H1N1鸡胚尿囊液(血凝滴度为1∶320)滴鼻感染小鼠,建立肾阳虚小鼠外感模型。感染1 d后给分别给MXF和利巴韦林药物干预,连续给药6 d,每日1次,观察并记录小鼠的体质量、肛温变化,死亡数和死亡时间,比较各脏器指数变化。结果肾阳虚外感模型建立成功,肾阳虚模型建立成功后,小鼠死亡多发生在病毒感染后的2~5 d之内;与正常组比较,其余6组小鼠体质量显著降低(P〈0.01);肾阳虚外感组及肾阳虚外感利巴韦林组的肛温明显降低(P〈0.01),肾阳虚外感组及肾阳虚外感利巴韦林组的肺指数明显升高(P〈0.05)。与肾阳虚外感组比较,肾阳虚外感MXF组小鼠死亡率、肺指数均有所降低,并且小鼠肛温显著回升(P〈0.05);与正常外感组比较,正常外感MXF组小鼠死亡率、肺指数均有所降低,肛温也出现回升趋势;H1N1感染小鼠,对照组无死亡,正常外感组死亡率10%,肾阳虚外感组死亡率20%,利巴韦林分别给药干预后死亡率分别是30%和10%,MXF给药干预后正常外感组和肾阳虚外感组均无死亡现象,与正常组和肾阳虚外感模型组比较,MXF有降低小鼠死亡率的趋势。结论 MXF更适合于肾阳虚外感病证,为临床上方证相关用药提供了一定的实验依据。Objective: To investigate the difference in intervention of Mahuang Xixin Fuzi decoction on normal mice and Kidney-yang deficiency mice model with influenza virus. Methods: KM mice were randomly divided into six groups by weight and anus temperature ,including the normal group. The Kidney-yang deficiency mice model infected with influenza virus group and intervened by ribavirin or MXF group and so on, according to the weight and rectal temperature. Kidney-yang deficiency model mice were established by intraperitoneal injection of estradiol benzoate (8 mg/kg). Mice were infected with influenza virus H1N1 allantoic fluid (Hemagglutination titer 1:320) by intranasal to establish the mice model of influenza disease with kidney-yang deficiency. After infection the mice were treated for 6 days. Body weight and rectal temperature changes ,mortality and survival time were observed, comparing the visceral index. Results: After the model established, the infected mice died in 2~5 days. Compared with the normal group,the weight of other groups decreased significantly (P〈 0.01 );the Kidney-yang deficiency mice model infected with influenza virus group and intervened by ribavirin decreased significantly (P〈 0.01 ),and their lung index decreased (P〈 0.05). Compared with the normal group,the other index of mice inter- vened by MXF became normal. After infected with H1N1 ,there were no death in normal group;the death rate of the normal infected with influenza virus was 10% ;the Kidney-yang deficiency mice model infected with influenza virus was 20% ;the group intervened by ribavirin was 30% and 10% ;and there were no death between the group intervened by MXF,which indicated that the MXF could decrease the death rate of mice. Conclusion: MXF has better intervention for the Kidney-yang deficiency mice model infected with influenza virus, providing a certain reference value for the clinical combination of traditional Chinese medicine.
