摘 要:目的观察丹参对单侧输尿管结扎(UUO)模型大鼠肾脏TGF-β_1及MCP-1表达的影响,初步探讨其抗肾间质纤维的机理。方法应用单侧输尿管结扎致肾间质纤维化大鼠模型,SD雄性大鼠120只随机分为空白组,模型组,丹参高、中、低剂量组,吡格列酮组,每组20只,从模型建立前48 h开始灌胃,于造模后第7天,留取梗阻侧肾脏,采用免疫组化方法检测肾组织TGF-β_1及MCP-1的表达,RT-PCR方法检测TGF-β_1及MCP-1 mRNA的表达。结果与空白组相比较,UUO模型组中TGF-β_1、MCP-1表达明显增高,丹参高、中剂量组TGF-β_1、MCP-1表达水平明显低于UUO模型组(P<0.05或P<0.01)。结论丹参可通过抑制UUO模型大鼠肾组织TGF-β_1、MCP-1蛋白及其m RNA的表达,从而起到肾脏保护和抗肾间质纤维化的作用。Objective: To investigate the effect of Salvia miltiorrhiza on the expression of monocyte chemoat- tractant protein-1 (MCP-1) and transforming growth factor-β1 (TGF-β1) in the rats with unilateral ureterai ob- struction (UUO) and evaluate the protective effect of Salvia mihiorrhiza against renal interstitial fibrosis. Meth- ods: 120 male Sprague-Dawley rats were divided into 6 groups: the normal control group,the model group,the Salvia mihiorrhiza group (low dose,middle dose,high dose) and Piloglitazone group,20 rats in each group. 48 hours before the modeling,the rats received intragastric administration. On 7th day after modeling,side kidney was taken,and TGF-β1 and MCP-1 were detected with Immunohistochemical method and their mRNA expres- sions were detected with the method of RT-PCR. Results: The levels of TGF-β and MCP-1 expression were ob- viously higher than those in the normal control group, and compared with the model group ,it obviously reversed these changes in middle dose and high dose of Salvia mihiorrhiza. Conclusions: Salvia mihiorrhiza can decrease the expression of TGF-β1 and MCP-1 in the UU0 rats, and reduce the extent of renal fibrosis.
