摘 要:本研究采用PCR-DGGE技术分析长角血蜱的中肠菌群结构。无菌条件下采集长角血蜱中肠内容物,以细菌基因组DNA提取试剂盒提取内容物细菌总DNA,以通用引物扩增制备16S r DNA V3区,DGGE电泳分离后,切胶回收、测定序列。结果表明,长角血蜱饱血雌成蜱中肠优势菌为泛菌属、柯克斯氏体属、肠杆菌属、绿脓杆菌属、假单胞菌属、克雷伯氏菌属、欧文氏菌属,但半饱血雌成蜱和雄成蜱中肠未检测到柯克斯氏体属(Coxiella)。The bacteria in the midgut of H.longicornis were detected with PCR-DGGE. The midgut's contents were collected under sterile conditions, and the bacterial total genomic DNA was extracted. Then the 16 S rDNA V3 was amplified with universal primers, and the DNA fragements were separated with DGGE electrophoresis. The result showed that Pantoea sp, Coxiella sp, Enterobacter sp, Pseudomonas aeruginosa sp, Pseudomonas sp, Klebsiella sp, Erwinia sp were dominant bacterial groups in the midgut from engorgement female ticks, but Coxiella sp was not found in the feding female and male H.longicornis.
