快速起搏对家兔心房肌钙通道表达的影响及辛伐他汀的干预研究

摘　　要：目的通过建立快速起搏家兔模型,探讨辛伐他汀对快速起搏早期心房肌细胞L型钙离子通道α1C亚基信使RNA(mRNA)表达的影响。方法 30只家兔按随机数字表法分为对照组(n=10),心房快速起搏组(n=10)和辛伐他汀干预组(n=10)三组。辛伐他汀干预组予以辛伐他汀5 mg/(kg·d)灌胃2周,其余组以等量生理盐水灌胃2周,对照组不行起搏,心房快速起搏组和辛伐他汀干预组行快速心房起搏(800次/min)建立家兔快速心房起搏模型,急性起搏8 h后取右心房组织,应用半定量反转录-聚合酶链反应检测心房肌L型钙离子通道α1C亚基mRNA的表达。结果对照组L型钙离子通道α1C亚基mRNA的相对拷贝数为1.00±0.04,辛伐他汀干预组为0.89±0.04,心房快速起搏组为0.78±0.03。心房快速起搏组和辛伐他汀干预组L型钙离子通道α1C亚基mRNA的相对拷贝数均显著低于对照组(P<0.05),心房快速起搏组L型钙离子通道α1C亚基mRNA的表达水平低于辛伐他汀干预组(P<0.05)。结论辛伐他汀可以改善家兔快速起搏心房引起的心房肌L型钙离子通道α1C亚基mRNA表达的下降。Objective To investigate the effect of simvastatin on mRNA expression of atrial α1c subunit of L-type calcium channel on atrial remodeling through establishing rapid atrial pacing rabbit models.Methods Total of 30 rabbits were divided into 3 groups according to random number table method： control group（ n = 10）,rapid pacing group（ n = 10） and simvastatin group（ n = 10）. Simvastatin 5 mg /（ kg·d） was given intragastricaly daily for two weeks before electrophysiology study in simvastatin group,normal saline was given intragastricaly in control and rapid pacing group instead. Control group with no pacing,while in simvastatin group and rapid pacing group,right atrium was paced at 800 beats / min for 8 hours to establish acute atrial fibrillation models,right atrium tissue was obtained for measurement of mRNA expression of α1c subunit of L-type calcium channel by reverse transcription-polymerase chain reaction（ RT-PCR）. Results In the control group,the relative copy number of α1c subunit mRNA of L-type calcium channel was 1. 00 ±0. 04; 0. 89 ± 0. 04 in simvastatin group,0. 78 ± 0. 03 in rapid atrial pacing group. The relative copy number of α1C subunit mRNA of L-type calcium channel in rapid pacing group and simvastatin group were significantly lower than the control group（ P〈0. 05）,rapid pacing group lower than simvastatin group（ P〈0. 05）.Conclusion Pretreatment with simvastatin can improve the degree of mRNA expression of α1c subunit of L-type calcium channel in rapid atrial pacing rabbit models.
