摘 要:目的探讨MF59佐剂对肠道病毒71型(enterovirus 71,EV71)灭活疫苗诱导小鼠体液免疫应答的影响。方法将ICR小鼠随机分成10组:MF59佐剂+不同含量EV71灭活抗原(50 EU、100 EU)组、Al(OH)3佐剂0.5 mg+不同含量EV71灭活抗原(50 EU、100 EU)组和PBS阴性对照组,每组10只,分别经小鼠后肢肌肉和腹腔注射,200μl/只,共免疫2次,间隔4周(0、28 d)。分别于初免和加强免疫后28 d,采集小鼠尾静脉血,分离血清,采用微量中和试验法检测小鼠血清抗体水平。结果除阴性对照组外,各组小鼠血清抗体阳转率及中和抗体GMT值均随免疫剂量和免疫时间的延长呈升高趋势。初免后28 d,抗原含量为50 EU(低剂量)和100 EU(高剂量)各组中和抗体GMT均较低,组间差异均无统计学意义(P>0.05)。加强免疫后28 d,低剂量各组小鼠血清中和抗体GMT与初免比较均略有升高(P>0.05);高剂量各组小鼠血清中和抗体GMT与初免比较均明显升高,其中MF59佐剂肌肉注射组血清中和抗体GMT值最高(274.40),显著高于铝佐剂肌肉注射组(P<0.05)。结论 MF59佐剂可显著增强EV71灭活疫苗诱导小鼠的体液免疫应答。Objective To investigate the effect of MF59 adjuvant on humoral immune response induced by inactivated enterovirus 71 vaccine in mice. Methods ICR mice were randomly divided into ten groups, ten for each, and inoculated twice with MF59 adjuvant + inactivated EV71 vaccine at dosages of 50 and 100 EU, 0. 5 mg aluminum hydroxide adju-vant + inactivated EV71 vaccine at dosages of 50 and 100 EU and PBS as negative control, each in a volume of 200 μl,by i. m. and i. p. routes, respectively, at an interval of 4 weeks(on days 0 and 28). Serum samples were collected 28 d after the first and the second injection respectively, and determined for antibody level by micro-neutralization test. Results Except those in negative control groups, both the antibody positive conversion rates and GMTs of neutralizing antibody in sera of mice in various groups showed increasing tendencies with the increasing dosage and time. The GMTs of neutraliz-ing antibody in the mice 28 d after the first injection with vaccine at dosages of 50 and 100 EU were relatively low, which showed no significant difference in various groups(P〉0. 05). However, 28 d after the second injection, the GMTs of mice injected with vaccine at a dosage of 50 EU increased slightly(P〉0. 05), while those at 100 EU increased significantly, as compared with those after the first injection. The GMT in MF59 adjuvant + 100 EU vaccine group was the highest(274. 40), which was significantly higher than that in the mice injected i.m. with the vaccine at the same dosage using aluminum hydroxide adjuvant(P〈0. 05). Conclusion MF59 adjuvant enhanced the humoral immune response induced by inactivated EV71 vaccine in mice.
