摘 要:目的:研究氧化白藜芦醇在体外对肿瘤细胞株以及小鼠移植性肉瘤S180的抑制作用。方法:采用四甲基偶氮唑蓝(MTT)法检测氧化白藜芦醇对人的各种肿瘤细胞株体外增殖的抑制作用,并计算其半数抑制浓度(IC50)。建立小鼠S180肉瘤移植瘤模型,分为对照组、环磷酰胺组、氧化白藜芦醇80、40、20mg·kg-1剂量组,以实体瘤瘤块重量及抑瘤率为指标评价氧化白藜芦醇对小鼠移植瘤生长的抑制作用,并计算其胸腺指数和脾脏指数。结果:MTT结果显示,氧化白藜芦醇对K562,SK-OV-3,MCF-7,SMMC-7721,A549,HNE-1和SW-480细胞株IC50值分别为52.88±3.21,11.55±5.42,23.61±2.13,58.13±4.84,7.44±2.94,67.73±7.23和62.86±5.2μg·ml-1,有较好的抑制肿瘤细胞体外生长作用。体内试验显示氧化白藜芦醇对移植瘤的抑制作用明显,80、40、20mg·kg-1三个剂量组对S180移植瘤抑制率为77.78%,61.43%和38.56%,呈一定量效关系,并对胸腺指数和脾脏指数影响不大。Objective:To investigate the inhibitory effect of oxyresveratrol on cancer cell line and sarcoma S180 of mouse.Methods:MTT assay was used to detect the inhibitory effect of oxyresveratrol on human cancer cell line proliferation in vitro.Sarcoma S180 of mouse model was performed to observe the anti-cancer activity of oxyresveratrol in vivo.Animals were divided into five groups including:negative control group,CTX group and three groups treated with oxyresveratrol at the dose 80,40,20mg·kg-1.The inhibitory effect of oxyresveratrol in vivo was evaluated by tumor size and inhibitory rates.Results:MTT assays showed that the IC50 of oxyresveratrol for the K562,SK-OV-3,MCF-7,SMMC-7721,A549,HNE-1and SW-480 were 52.88±3.21,11.55±5.42,23.61±2.13,58.13±4.8,7.44±2.94,67.73±7.23和 62.86±5.2μg·ml-1respectively.Oxyersveratrol of 80,40,20mg·kg-1 had significant inhibitory effects and dose-dependent effects on growth of tumor,the inhibitory rates of tumor were77.78%,61.43%(P〈0.01),61.43%(P〈0.05)and 38.56%.Oxyresveratrol have obvious anti-tumor activities both in vivo and in vitro.
