摘 要:目的:建立左归丸提取液中多糖的含量测定方法,优选左归丸水提醇沉法最佳醇沉浓度.方法:以毛蕊花糖苷转移率、多糖转移率、干膏得率为指标,采用HPLC-VWD 测定毛蕊花糖苷含量,采用苯酚-硫酸比色法测定多糖含量,干燥失重测定法测干膏得率,以多指标综合评分法优选水提醇沉浓度.结果:所建立的多糖含量测定方法,线性范围为2.53~12.65μg,平均回收率为96.98%(RSD为3.56%).醇沉浓度30%、50%、70%时,毛蕊花糖苷转移率分别为81.38%、90.28%、98.60%,多糖转移率分别为50.77%、46.37%、20.69%;醇沉浓度为50%时,保留大部分多糖和毛蕊花糖苷,为最佳醇沉浓度.结论:建立的多糖含量测定方法简单易行、重复性良好.醇沉浓度对毛蕊花糖苷、多糖转移率影响均较大,对干膏得率影响不大.Objective:To establish methods for determination of the polysaccharide in the extracting solution of Zuogui pills and optimize the concentrations of alcohol in precipitation method of zuogui pills and.Methods:The polysaccharide transfer rate,acteoside transfer rate and the yield of dry extract was used as index components,the content of the acteoside was detected by the HPLC-VWD method;The phenol-sulfuric acid method was adopted to determine the content of the polysaccharide,the loss on drying method was used to detect the yield of dry extract;The best concentration of alcohol in precipitation method was optimized by the above extraction process.Results:The linear range of polysaccharide was 2.53~12.65μg,the average recovery rates was 96.98%(RSD was 3.56%).When the alcohol precipitation concentration of 30%,50%,70%,the acteoside transfer rate were 81.38%,90.28%,98.60%,the polysaccharide transfer rate were 50.77%,46.37%,20.69%.when the alcohol precipitation concentration was 50%,it would transfer most of the acteoside and the polysaccharide.Above all,The best concentration of alcohol in precipitation method is50%.Conclusion:The content determination method was simple and feasible with good reproducibility,the alcohol precipitation concentration has great impact on the polysaccharide transfer rate and acteoside transfer rate,but slight influence on the yield of dry extract.
