摘  要:目的:研究不同宫内缺氧时间对胚胎大鼠脑组织信号转导子与转录激活子3蛋白表达的影响。方法:制备乏氧性缺氧动物模型,从胎龄15d到16d连续宫内缺氧2d,分别缺氧2、4、6、8、12h共5个不同时程,采用免疫组化检测胚胎大鼠脑组织STAT3蛋白表达情况。结果:与对照组比较,缺氧2h胎鼠脑组织STAT3蛋白表达升高(P<0.05),在缺氧6h达最高(P<0.05);缺氧8h较对照组染色增强,但较缺氧6h表达减弱(P<0.05);缺氧12h组与对照组相比STAT3表达降低(P<0.05)。结论:短时缺氧(缺氧2、4、6、8h)能增强胎鼠脑组织STAT3蛋白的表达,但缺氧时间较长(缺氧12h)会降低STAT3蛋白的表达,说明宫内缺氧对胚胎脑组织的影响可能通过STAT3通路进行调节。Objective To study the effect of intrauterine hypoxia in different time on the expression of STAT3 in rat embryo Brain tissue. Methods C, estational age 15 to 16 days rats were used of intrauterine hypoxia animal models. And the time of hypoxia respectively were 2, 4, 6, 8, 12h in a day. The protein expression levels of STAT3 in the Brain tissue of rat embryos were detected by im- munohistochemical. Results By comparing with the control group, the protein expression levels of STAT3 in the Brain tissue of rat embryos were increased from hypoxia 2h, reached the highest level at 6h and decreased at 12h (P 〈 0. 05). Conclusion Hypoxia 2, 4, 6h and 8h can enhance the expression of STAT3 in the Brain tissue of rat embryos. But hypoxia 12h reduce the expression of STAT3. STAT3 possibly plays an important action in Hypoxic isehemia encephalopathy.
