摘 要:目的优化鹅藻滴鼻剂鹅不食草的提取纯化工艺。方法采用紫外分光光度法,单因素试验,以乙醇浓度、料液比、提取时间、提取次数为因素进行L9(34)正交试验,以总黄酮的提取率,纯化提取转移率为考察指标,优选鹅不食草最佳提取纯化工艺技术参数。结果最佳提取工艺条件为以药材14倍量的35%乙醇提取3次,每次1.5 h,总黄酮的提取率为75.93%;提取浓缩液(浓缩液∶药材=1∶1)以70%醇沉,总黄酮的提取转移率为63.07%。结论本提取纯化工艺可操作性强,提取转移率高,适用于鹅藻滴鼻剂的研发。OBJECTIVE To optimize the extraction and purification technology of the herba centipedae for E'zao nasal drop.METHODS UV spectrophotometry was adopted to detect total flavonoids and single factor tests are employed. The factors such as ethanol concentration,dosage liquor ratio,extraction time and number of times was used to carry out L9( 34) orthogonal test. Total flavonoids extraction percentage and purification extraction diversion rate were employed to inspect and optimize the best technical parameter of herba centipedae's extraction and purification technology. RESULTS The optimal extraction technology was as follows:35% ethanol as 14 times of drugs was used to extract total flavonoids for 3 times and every time for 1.5 h,and the extraction rate was75.93%. The extracted concentrated solution( concentrated solution: drug = 1 ∶ 1) was precipitated by 70% ethanol,and the total flavonoids diversion rate was 63.07%. CONCLUSION This maneuverability of extraction and purification technology is feasible with high extraction efficiency,and it could be applied to research and develop the E'zao nasal drop.
