摘  要:采用过氧化氢降解法制备水溶性壳聚糖,分别对过氧化氢质量分数、反应温度、反应时间、乙酸质量分数四个因素进行了研究,采用外推法测定低聚壳聚糖的分子量,红外光谱法确定其结构。通过正交实验得到过氧化氢降解壳聚糖的最优条件为:H_2O_2浓度4%,反应时间4h,反应温度60℃,乙酸浓度4%,摇床转速150r/min,此条件下的低聚壳聚糖回收率为32%,粘均分子量为4566.9。经红外光谱分析,制备的水溶性壳聚糖与其原料壳聚糖结构一致。The preparation of soluble chitosan through the degradation of chitosan by H202 , the factors affecting the chitosan -degrading such as its mass fraction, reaction temperature, reaction time, the mass fraction of hydrogen peroxide, the mass fraction of acetic acid were studied and the conditions of the degradation were optimized. The degradation was determined by extrapolation of chitosan molecular weight. IR spectroscopy was used to determine its structure. By orthogonal experiment of hydrogen peroxide degradation of chitosan, the optimal conditions were: reaction time 4h, reaction temperature 60℃, H2O2 concentration of 4% , acetic acid concentration of 4%. Under this condition, the yield of chitosan oligomer was 32%, viscosity - average molecular weight was 4566.9. Characterized by IR spectrum analysis, it was known that the structure of chitosan oligomer was the same as the raw material chitosan.
