摘  要:目的研究分泌型白细胞蛋白酶抑制因子(SLPI)对脂多糖(LPS)诱导的人肺上皮细胞株A549中肺表面活性物质D(SP-D)表达的影响。方法体外培养A549细胞,分为空白对照组(A组)、LPS 10μg/ml刺激组(B组)、pcDNA3.1(+)-SLPI转染组(C组)和pcDNA3.1(+)转染组(D组)。采用RT-PCR和Western blot分别检测SP-D mRNA和蛋白表达。结果与A组相比,C组SP-D mRNA和蛋白表达增加(P<0.05),而B组和D组SP-D mRNA和蛋白表达降低(P<0.05);C组SP-D mRNA和蛋白表达均较B组增加(P<0.01)。结论 SLPI能拮抗LPS的炎性作用,刺激人肺上皮细胞中SP-D的分泌,提示SLPI对LPS诱导的肺损伤具有保护作用。Objective To inve,;tigate the effect of secretory leukocyte protease inhibitor(SLPI) on pulmonary surfactant protein D(SP-D) expression induced by lipopolysaccharide (LPS) in human pulmonary epithelial cell line A549. Methods A549 cells were cultured in vitro and then divided into four groups of A(blank control), B(stimulated with LPS 10 Effect of SLPI on SP-D expression induced by lipopolysaccharide in human pulmonary epithelial cellsg/ml), C[transiently transfected with pcDNA3. 1 (+)-SLPI plasmid after LPS stimulation] and D [ transiently transfected with pcDNA 3.1(+) plasmid after LPS stimulation]. The mRNA and protein expressions of SP-D were detected by RT-PCR and Western blot, respectively. Results Compared with group A, the mRNA and protein expressions of SP-D were increased in group C(P〈0. 05) ,which were decreased in groups of B and D (P〈0. 05). Moreover, the mRNA and protein expressions of SP-D in group C were higher than those in group B (P〈0. 01 ). Conclusion SLPI can antagonize the inflammatory response of LPS and promote SP-D secretion in A549 cells,which indicates that SLPI has a protective effect on LPS-induced lung injury.
