摘 要:拟南芥2-氧化戊二酸依赖的双加氧酶基因F6’H1缺失突变体f6’h1出现叶片衰老延迟表型。利用根癌农杆菌介导的花序浸染法转化拟南芥,再以抗性筛选方法获得了F6’H1基因诱导过量表达拟南芥植株,地塞米松(DEX)喷施处理24 h能显著诱导F6’H1基因上调表达,F6’H1基因的过量表达使转基因拟南芥叶片衰老提前。结果表明,拟南芥2-氧化戊二酸依赖的双加氧酶基因F6’H1正向调控叶片衰老。2-oxoglutarate dependent dioxygenase gene F6'H1 knockout mutant f6'h1 exhibited significantly delayed leaf senescence in Arabidopsis thaliana.A dexamethasone(DEX,a synthetic glucocorticoid) inducible overexpression system containing the F6'H1 gene was transformed into wild-type plants through floral dipping method,which was mediated by Agrobacterium tumefaciens.The transgenic plants resistant to hygromycin and kanamycin were obtained.The transcription of F6'H1 in transgenic leaves was increased after DEX spraying whole plant.Comparing with the wild-type plants,the precocious leaf yellowing were observed 5 d after the DEX treatment in F6'H1 inducible overexpression lines.Taken together,2-oxoglutarate dependent dioxygenase gene F6'H1 positively regulate leaf senescence of Arabidopsis thaliana.
