摘  要:目的体外观察山银花黄酮粗提物对MCF-7细胞增殖和凋亡的影响,为其黄酮类化合物的进一步开发利用提供理论依据。方法 MTT法检测山银花粗提物对MCF-7细胞增殖的抑制作用;Hoechst33342染色法观察MCF-7细胞凋亡;流式细胞术检测MCF-7细胞的凋亡率。结果高于200μg·m L-1的山银花粗提物对MCF-7细胞增殖有显著抑制作用(P<0.05)。山银花粗提物对细胞形态有明显影响,随浓度的增加,细胞核被染成亮蓝色的细胞比例明显增加。0μg·m L-1、100μg·m L-1、300μg·m L-1、600μg·m L-1的山银花粗提物作用的MCF-7细胞凋亡率分别为(3.89±0.70)%、(10.40±0.92)%、(22.02±1.22)%、(27.03±0.75)%。结论山银花黄酮粗提物对MCF-7肿瘤细胞有明显的抑制增殖与诱导凋亡作用,其效应与浓度呈正相关。Objective To investigate the effects of flavoniods crude extract from Flos Lonicerae Confusae on the prolif- eration and apoptosis of MCF-7 cells, so as to provide the material foundation and scientific basis for further exploration and utilization. Methods The inhibition effects on proliferation of MCF-7 cells were tested by MTT. The morphological changes of MCF-7 apoptosis were observed by Hoechst 33342 staining. Flow cytometry was involved to measure the apoptosis rate of MCF-7 ceils. Results The flavoniods crude extract of concentration more than 200 μg·m L-1 had great inhibition effect on the proliferation of MCF-7 cells (P 〈 0.05 vs. control). The crude extract also had a significant effect on cell morphology. The cell ratio of nucleus stained into hght blue increased along with the increase of concentration of the exlracts. The apoptosis rates of MCF-7 induced by a concentration of 100μg·m L-1, 300μg·m L-1 and 600μg·m L-1 were (10.4 ±0.92)%, (22.02 ±1.22)% and (27.03 ± 0.75)%, respectively. Conclusion The crude flavonoids extracts from Flos Lonicerae Confusae had great inhibition on proliferation and induction to apoptosis of MCF-7 cells. The effect was positively correlated with its concentration.
