摘  要:目的:研究天芪益智颗粒对快速老化小鼠学习记忆能力和脑组织内中枢神经递质的影响。方法:以自然老化小鼠SAMR 1为对照组,将快速老化小鼠SAMP 8随机分为拟阿尔茨海默病(Alzheimer's disease,AD)模型组、石杉碱甲组、天芪益智颗粒高、中、低剂量组,每组10只,连续灌胃给药60 d。实验结束次日,采用筑巢实验、避暗实验和Morris水迷宫测试各组小鼠的学习和记忆能力。采用高效液相色谱法检测各组小鼠脑组织中去甲肾上腺素(NE)和5-羟色胺(5-HT)的含量。结果:与对照组比较,模型组小鼠筑巢实验得分明显降低(P<0.05);避暗实验的步入潜伏期显著缩短(P<0.01),错误次数明显增多(P<0.05);水迷宫定位巡航实验的逃避潜伏期明显延长(P<0.05),空间探索实验的穿越目标象限平台次数显著减少(P<0.01)。与模型组比较,天芪益智颗粒中剂量组的筑巢实验得分明显增高(P<0.05);高剂量组的避暗实验步入潜伏期显著延长(P<0.01),错误次数明显减少(P<0.05);高、中剂量组在空间探索实验中穿越目标象限平台的次数明显增多(P<0.01,P<0.05);中剂量组在水迷宫定位巡航实验中的逃避潜伏期明显缩短(P<0.05),同时,天芪益智颗粒组脑组织中的NE和5-HT含量明显升高(P<0.05)。结论:天芪益智颗粒能够改善快速老化小鼠的学习和记忆能力,其机制可能与调节脑组织内中枢神经递质NE和5-HT的含量相关。Objective: To study the effect of Tianqi Yizhi Granule on senescence accelerated mice learning and memory ability and brain neurotransmitters. Methods: SAMR1 natural aging mice were as a control group and rapid aging mice SAMP 8 were randomly divided into Alzheimer's disease( Alzheimer's disease,AD) model group,Huperzine A group,Tianqi Yizhi Granules high,medium and low dose groups,10 rats in each group,continuous intragastric administration for 60 days. On the next day of the experiment ending,by using nest experiment,passive avoidance learning and memory test and Morris water maze were used to test mice in each group. We detected the mice brain norepinephrine( NE) and serotonin( 5- HT) content using high performance liquid chromatography. Results: Compared with the control group,the experimental model group's nest score was significantly lower( P〈0.05). The step- through latency of passive avoidance test was significantly shorter( P〈0.01). The number of errors was increased significantly( P〈0.05). Water maze experiment positioning cruise escape latency was significantly longer( P〈0.05).The target quadrant frequency of space exploration experiment platform was significantly reduced( P〈0.01). Compared with the model group,nest experiment's score of Tianqi Yizhi Granule groups was significantly higher( P〈0.05). The dark experiment's avoidance step- through latency of high- dose group was significantly prolonged( P〈0.01). The number of errors was decreased obviously( P〈0.05). High and medium dose groups' target quadrant number of experiment platform in space was significantly increased( P〈0.01,P〈0.05). Medium dose group' s escape latency in water maze experiment was significantly shorter( P〈0.05). Meanwhile,NE and 5- HT contents of Tianqi Yizhi granules in brain tissue were significantly higher( P〈0.05).
