红芪多糖3促进小鼠脾淋巴细胞增殖的差异蛋白质点分析

摘　　要：目的:分析红芪多糖3(HPS-3)作用于小鼠脾淋巴细胞后的差异蛋白质点,探讨红芪多糖3调节免疫的分子生物学机制。方法:提取正常小鼠脾淋巴细胞进行体外培养,用MTT法检测红芪多糖对小鼠脾淋巴细胞的刺激增殖作用,ELISA试剂盒测定培养上清中的IL-2含量,并运用双向电泳技术对脾淋巴细胞总蛋白进行分离,银染显色,PDQuest软件对凝胶图谱进行差异点分析,找出差异明显的蛋白质点。结果:HPS-3能够提高T淋巴细胞增殖能力和IL-2的分泌,实验所得2-DE凝胶图谱经PDQuest软件分析得出以下结果:HPS-3组中共有16个蛋白点发生明显改变,其中11个蛋白质点高表达,4个蛋白质点消失,1个蛋白质点低表达。结论:HPS-3能促进脾淋巴细胞增殖,显著影响小鼠脾淋巴细胞蛋白质表达。Objective： To study the differential protein spots expression in mice spleen lymphocytes treated wth Radix Hedysari Polysaccharides（ HPS- 3）. Methods： HPS- 3 was given to the mice spleen lymphocytes and the comparison was made with the proliferation index of T lymphocyte and the level of IL- 2 in the cultural supernatants. The two- dimentional gel electrophoresis（ 2- DE） was used to isolate the total proteins extracted from the mice spleen lymphocytes,and then the 2- DE gels were stained by silver nitrate. PDQuest software was used to analyze the gel maps and we found out differential protein spots. Results： HPS- 3can improve the ability of T lymphocyte proliferation and secretion of IL- 2. Analysis results of the 2- DE gel maps with PDQuest software： there were 16 spots changed significantly in HPS- 3 group,including eleven up- regulated and one down- regulated and four disappearing. Conclusion： HPS- 3 can promote the mice spleen lymphocytes proliferation and increase the level of IL- 2and dramatically impacts the expression of the proteins mice spleen lymphocytes.
