摘  要:目的探讨持续性高正加速度(+G_z)对大鼠舌横纹肌组织结构、热休克蛋白70(heat shock protein 70,HSP70)m RNA表达的影响,以及对丹参多酚酸盐的作用。方法将27只SD大鼠随机分为9组:阴性对照组、+5 G_z阳性对照组、+5 G_z药物组(低、中、高剂量)、+10 G_z阳性对照组、+10 G_z药物组(低、中、高剂量),每组3只。阳性对照组和药物组在+5 G_z、+10 G_z条件下离心,给药组于每次离心前30 min腹腔注射不同浓度的丹参多酚酸盐溶液,其余组注射等体积生理盐水。观察舌横纹肌组织形态,荧光定量PCR法测定HSP70 m RNA表达情况。结果相对于阴性对照组,其他组舌横纹肌组织纤维基本正常,偶见细胞间质稀疏,细胞排列紊乱及横纹不清或消失;阳性对照组HSP70 m RNA的表达高于阴性对照组,且随+G_z值的增加而增加;药物组HSP70 m RNA的表达低于阳性对照组。结论 +G_z对大鼠舌横纹肌可造成一定的损伤,损伤程度与+G_z值相关;丹参多酚酸盐可抑制+G_z作用下HSP70 m RNA的表达,对+G_z暴露下舌组织可能有一定保护作用。Objective To investigate the possible inf luence on high +Gz exposure on structure and heat shock protein 70(HSP70)m RNA relative expression in tongue striated muscle of SD rats, and to observe the effects of depside salt from salvia miltiorrhiza on high +Gz exposure. Methods A total of 27 male SD rats were randomly divided into 9 different groups: negative control group, +5 Gz positive control group, +10 Gz positive control group, +5 Gz positive treatment groups(high, middle and low dose), +10 Gz positive treatment groups(high, middle and low dose), with 3 rats in each group respectively. The rats in positive control and dr ug groups were f ixed with rats kept-keeling devices for centrifugation exposed to +5 Gz and +10 Gz, the rats in drug groups were injected into abdomen with different doses depside salt from salvia miltiorrhiza 30 min prior to +Gz exposure, the rats in other groups were injected with the same value normal saline. The tongue tissues were observed by histological method through light microscope, and real time q PCR was applied to determine the relative expression of HSP70 m RNA. Results The normal structure of tongue striated muscle was observed in the negative control group, fibers of striated muscle for other groups were basically normal, with sparse for intercellular substances occasionally, cell ar rangement disorder and muscle text ure indistinctly. Compared to the negative cont rol group, the relative expression of HSP70 m RNA significantly higher and increased with the trend of raising +Gz value in positive control groups. Compared to the positive control groups, the relative expression of HSP70 m RNA decreased in the treatment groups. Conclusion +Gz may cause injury to the tongue striated muscle through increasing relative expression of HSP70 m RNA, and the injury extent was related to +Gz raising. Depside salt from salvia miltiorrhiza can inhibit HSP70 m RNA expression on +Gz exposure and have protective effects on tongue tissue on +Gz conditio
