摘  要:目的建立HPLC同时测定鸡眼草药材中染料木素和山柰酚含量的方法。方法采用RPHPLC法。色谱柱为Diamons C_(18)(150 mm×4.6 mm,5μm),流动相为甲醇-体积分数0.1%磷酸水溶液(体积比为72∶28),流速为1.0 m L/min,检测波长为370 nm,柱温为30℃。结果染料木素和山柰酚质量浓度分别在0.50~4.02 mg/L和0.15~1.20 mg/L内与峰面积呈良好的线性关系,平均回收率分别为99.7%和100.9%,RSD分别为0.1%和0.7%(n=6)。结论建立的方法简便,准确,重现性好,可用于鸡眼草药材中染料木素和山柰酚的含量测定。Objective To establish a method for the simultaneous quantitative analysis of genistein and kaempferol in the aerial parts of Kummerowia striata with HPLC. Methods The chromatographic column was Diamonsil C_(18)( 150 mm× 4. 6 mm,5 μm); the mobile phase was methnol and aqueous solution containing 0. 1 % phosphoric acid with the volume ratio of 72∶ 28 and the flowrate was 1. 0 mL / min; the UV detection wavelength was 370nm; the oven temperature was 30 ℃. Results The linear ranges of genistein and kaempferol were 0. 50 ~ 4. 02 mg / L and 0. 15 ~ 1. 20 mg / L,respectively. The average recoveries( n = 6) of genistein and kaempferol were 99. 7% with RSD of 0. 1%,100. 9% with RSD of 0. 7%,respectively. Conclusion The method is simple and accurate,it can be used to determine genistein and kaempferol in the aerial parts of Kummerowia striata.
