隆林山羊ACSS2基因的克隆及序列分析

摘　　要：根据藏系绵羊的ACSS2基因序列设计克隆引物,并以隆林山羊肌间脂肪组织为模板克隆ACSS2基因并测序。利用在线生物信息学软件分析ACSS2蛋白的序列特性,分析不同物种间ACSS2基因的进化关系及三级结构,从而预测隆林山羊ACSS2基因的功能。结果发现,隆林山羊ACSS2基因CDS全长2 103bp,由18个外显子组成,编码701个氨基酸残基。与牛(NM_001105339)、人(NM_018677)、绵羊(XM_012114899)、小鼠(NM_019811)序列的ACSS2基因相比较,核苷酸相似性分别为97.9%、91.5%、97.6%和88.2%,氨基酸的相似度分别为98.4%、93.6%、97.4%和91.9%。其中绵羊较山羊多出13个氨基酸(G279-Q291,GKPKEKPKHIRPQ)。隆林山羊ACSS2蛋白与其他物种间具有相似的三级结构系统,且与绵羊和牛具有较近的亲缘关系,而与猪的亲缘关系较远。The primers were designed according to the ACSS2 sequence of Tibetan sheep,CDS of ACSS2 was cloned and sequenced by RT-PCR.Using online tools,the sequence character,structure prediction and evolution analysis were performed,which may be helpful for the function prediction of ACSS2 in Longlin goat.The results showed that the coding region of ACSS2 of Longlin goat is 2 103 bp,comprised of 18 exons,coding 701 amino acids.The similarity of RNA sequence and protein sequence between Longlin goat and bovine,human,sheep,mouse are 97.9%,91.5%,97.6%,88.2% and 98.4%,93.6%,97.4% and 91.9%respectively.The protein sequence of sheep ACSS2 is longer than other species including goat et al by 13 amino acids.The tertiary structure of goat showed high similarity with bovine,human and mouse,and has close genetic relationship with ovis and bovine,but a far distance with pig.
