复方“前愈”有效部位群的分离纯化及含量测定

摘　　要：目的从中药复方"前愈"中提取分离有效部位群并将其进行纯化,建立各有效部位(总生物碱,总黄酮,总多糖,总皂苷)的含量测定方法。方法采用溶剂法分离纯化总生物碱有效部位;溶剂法结合聚酰胺树脂柱层析法分离纯化总黄酮有效部位;水溶醇沉法分离纯化总多糖有效部位;采用溶剂法结合大孔树脂柱层析法分离纯化总皂苷有效部位。应用紫外分光光度法分别测定分离提取液及纯化液中各有效部位的含量。结果提取分离所得总生物碱,总黄酮,总皂苷,总多糖含量分别为2.07%,8.87%,9.46%,2.82%。纯化后各部位含量分别为50.52%,57.04%,74%,67.8%。结论本实验确定的分离纯化方法操作简便合理,可显著提高"前愈"复方中各有效部位的含量。Objective To extract,separate and purity the effective fractions of the traditional Chinese medicine＂Qianyu＂and establish the content determination method of the effective fractions（ total alkaloids,total flavonoids,total polysaccharides and total saponins）. Methods To separate and purificate the total alkaloids by solvent method; to separate and purificate the total flavonoids from the effective fraction by solvent method and polyamide resin column chromatography; to separate and purificate the total polysaccharide fractions by water soluble alcohol precipitation; to separate and purificate the total saponins from the effective fraction by solvent method and macroporous resin column chromatography. Determinate the content of active fractions in the extraction and purificated solution by ultraviolet spectrophotometry. Results The total polysaccharide content of the extraction and separation of the resulting total alkaloids,flavonoids,saponins were 2. 07%,8. 87%,9.46%,2. 82%. After purification,the content of each part were 50. 52%,57. 04%,50. 11%,67. 8%. Conclusion The experimentally determined separation and purification method is simple and reasonable,can significantly increase the content of each effective fraction of compound＂Qianyu＂.
