摘 要:目的:探讨乌骨藤总皂苷(saponins of Marsdenia Tenacissima,SMT)对小鼠急性肝损伤的保护作用及可能机制。方法:建立对乙酰氨基酚(APAP)诱导的小鼠急性肝损伤模型,分为正常组、模型组、SMT给药组(15,30和60 mg·kg-1),紫外-分光光度法检测各组血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)活性以及肝组织匀浆中超氧化物歧化酶(SOD)、丙二醛(MDA)水平,HE染色法观察肝脏病理学改变。结果:与模型组比较,SMT(30和60 mg·kg-1)灌胃给药能明显降低血清中升高的ALT,AST活性,同时发现SMT可降低肝组织匀浆中升高的MDA和SOD水平。SMT可明显改善肝组织坏死范围和程度,减少炎细胞浸润。结论:SMT对APAP所致急性肝损伤小鼠具有一定的保护作用,其机制可能与SMT抗氧化作用等有关。Objective: To determine the protective effect of saponins of Marsdenia Tenacissima( SMT) on acute liver injury induced by N-acetyl-p-aminophenol [N-( APAP) ]in mice. Methods: Acute liver injury was induced by APAP in mice. The mice were divided into 5 groups: normal group,model group,SMT dose groups( 15,30 and 60 mg·kg- 1). The serum activities of alanine aminotransferase( ALT) and aspartate aminotransferase( AST) were determined by ultraviolet spectroscopy. Simultaneously,superoxide dismutase( SOD) and malondialdehyde( MDA) contents in liver homogenates were also detected. Liver tissue sections were examined after HE staining. Results: In mice with APAP-induced acute liver injury,SMT( 30 and 60 mg·kg- 1) markedly decreased the elevated serum levels of ALT and AST. In addition,SMT significantly increased SOD activities,and decreased MDA content in liver homogenates. SMT effectively attenuated the tissue lesion of liver. Conclusion: SMT has the protective effect on acute liver injury induced by APAP in mice. The mechanism may be attributed to increasing anti-oxidation capacity.
