摘  要:目的优化超临界CO_2萃取洛伐他汀的工艺。方法以洛伐他汀萃取率为指标,采用单因素试验分别考察不同萃取压力、萃取温度、萃取时间及夹带剂用量对洛伐他汀萃取率的影响,再用正交试验进行条件优化。结果洛伐他汀萃取的最佳工艺条件:萃取温度55℃,萃取压力25 MPa,萃取时间100 min,夹带剂用量为2 000 ml/kg,最佳工艺条件下洛伐他汀萃取率可达97.17%;各工艺参数对萃取率的影响排序为:萃取压力>萃取温度>萃取时间>夹带剂用量。结论该工艺具有萃取率高、速度快、操作简便等优点,可用于洛伐他汀的提取。Objective To optimize the supercritical CO2 extraction technology for extraction of lovastatin. Methods The single factor test was used to investigate the effect of extraction pressure,extraction temperature,extraction time and agent of co-solvent on the extraction yield of lovastatin. Then the orthogonal experiment was used to optimize the extraction conditions. Results The optimized extraction conditions were as follows: extraction pressure of 25 MPa,extraction temperature of 55 ℃,agent of co-solvent of 2 000 ml / kg,and extraction time of 100 min. Under these conditions,the yield rate reached 97. 17%. The influence of various parameters on the extraction rate was extraction pressure,extraction temperature,extraction time,agent of co-solvent in order from the highest to the lowest priority. Conclusion This method is simple,rapid and has higher extraction yield,and it is suitable for the extraction of lovastatin.
