摘 要:目的研究75%酒精超声清洗对同种脱钙骨基质(DBM)结构及成骨性能的影响。方法制备2组DBM,A组(75%酒精超声清洗处理),B组(常规处理),通过HE染色观察脱钙骨基质结构变化。建立裸鼠异位诱导成骨试验模型,通过拍摄X线片观察成骨情况,进而行大体观察和组织形态学观察,并进行成骨反应的组织学评分。结果结构观察:A组和B组均可见骨胶原结构保持完整,无明显破坏,2组无差异。X线观察:术后当天,裸鼠后腿部为正常肌肉显影,术后28 d双侧均可见较亮的显影。大体观察:术后观察裸鼠活动自如,切口愈合良好,脱钙骨基质被肌肉包裹,无肌肉坏死。A、B组无差异。组织形态学观察:术后28 d,HE染色可见A组:植入部位炎症反应较小,肌肉无坏死,脱钙骨基质有少量降解,周围或中间空隙可见一些透明软骨细胞,软骨细胞活跃,并开始分泌形成软骨基质。脱钙骨基质周围甚至有新生骨基质形成,可见成熟的骨细胞。B组与A组组织形态学观察结果大体一致。经组织学评分显示,2组差异无统计学意义(P>0.05)。结论 75%酒精超声清洗30 min不会对同种脱钙骨基质结构及性能造成损失,可作为脱钙骨基质制备的一道程序。Objective To study the effect of 75% ethanol ultrasonic cleaning on the structure and osteoinduction properties of human demineralized bone matrix. Methods Two demineralized bone matrix were prepared, group A: by 75% ethanol ultrasonic cleaning process,group B: the conventional treatment group. The structural change of demineralized bone matrix was observed by HE staining. Nude mice heterotopic model was selected. X-rays were taken to observe bone formation, and then gross observation, histological examination, and osteogenic scores were carried out. Results Group A and group B were seen with intact collagen structure, there were no obvious damage or difference in the two groups. X-ray observation: 0 days,normal muscle graphs, after 28 days bilaterally lighter areas could be seen. Gross observation: postoperative nude mice showed normal activities, with incision healing well, demineralized bone matrix wass wrapped by muscle, no muscle necrosis.Histological examination: after 28 days, HE staining: Group A showed no inflammation, muscle without necrosis, a small amount of demineralized bone matrix degradation, around the middle of the gap some hyaline cartilage cells could be seen,cartilage cells secreted cartilage matrix formation. New bone matrix formation was observed, showing mature osteoblasts. Group A and group B was quite consistent with respect to the histological observations. Histological scores also showed that there was no significant difference(P 〉0.05). Conclusion Seventy five percent ethanol ultrasonic cleaning will not impair the structure and performance of human demineralized bone matrix, and thus it can be used as detergent or sterile solution in preparing demineralized bone matrix.
