摘  要:采用表面消毒法从健康芍药根部组织中分离获得48株内生细菌,菌落形态存在明显差异,且种群数量达到1.21×10^3cfu/g鲜重,表明芍药根部组织中存在大量内生细菌且类群丰富。从48株内生细菌中选择34株菌落形态差异较大的内生细菌菌株,利用16S核糖体DNA限制性片段长度多态性聚合酶链反应的方法进一步分析其种群多样性。根据酶切图谱的聚类分析结果,供试的34株内生细菌在100%的相似性水平上归为22类,表明芍药根部内生细菌的种群多样性。各类群代表菌株16S核糖体DNA的序列分析表明芍药根部内生细菌包含5个属(芽孢杆菌属、假单胞杆菌属、泛菌属、肠杆菌属和氨醇杆菌属)的14个种,其优势种群主要为芽孢杆菌和假单胞菌。48 endopytic bacteria have been isolated from surface-sterilized roots of healthy paeonia lactiflora,and these strains varied in colony morphologies. The endopytic bacterial population density could reach 1. 21 × 10^3 cfu / g fresh weight,which indicates that a great amount of endopytic bacteria are existing and has a rich population diversity in the paeonia lactiflora roots. The genetic diversity of 34 from the 48 endophytic bacteria with significant differences in morphology has been further studied by 16 S r DNA PCR-RFLP. In the level of 100% similarity,UPGMA of 16 S rDNA PCR-RFLP map shows that 34 isolates are grouped into 22 clusters,which presents abundant genetic diversity. Moreover,16 S rDNA sequence analysis of representative strains of 22 clusters proves that the endophytic bacteria in paeonia lactiflora roots consist of 14 species of the 5 genera including Bacillus,Pseudomonas,Pantoea,Enterobacter and Sphingobacterium,among which Bacillus and Pseudomonas are the dominant genera.
