补肾活血固齿方对小鼠颅顶前成骨细胞增殖活性及细胞周期影响的实验研究

摘　　要：目的:观察补肾活血固齿方对小鼠颅顶前成骨细胞(MC3T3-E1细胞)增殖活性及细胞周期变化的影响,探讨该方剂治疗牙周炎的作用机理。方法:MC3T3-E1细胞分别于5%、10%、15%、20%补肾活血固齿方含药血清,10%无药血清,10%胎牛血清中培养12、24、36、48、60、72 h,MTT法检测补肾活血固齿方对MC3T3-E1细胞增殖活性的影响。另外MC3T3-E1细胞分别于10%补肾活血固齿方含药血清、无药血清、胎牛血清中培养24、48、72 h,用流式细胞仪检测该方剂对MC3T3-E1细胞细胞周期的影响。结果:含药血清与胎牛血清、无药血清组相比,MC3T3-E1细胞增殖活性及细胞周期差异无显著性(P>0.05)。结论:补肾活血固齿方对MC3T3-E1细胞增殖活性及细胞周期无影响,其可能是通过促进该细胞向成骨细胞分化、促进牙槽骨的再生而发挥药物作用。Objective： to observe the effect of Kidney-tonifying Blood-activating and Tooth-astringing Formula on MC3T3-E1 multiplication activity and the cell cycle,and to explore the functional mechanism of the formula in treating periodontitis. Methods：MC3T3-E1 were cultured respectively in formula serum of 5%,10%,15% and 20%,10% pure serum,and 10% fetal bovine serum for 12,24,36,48,60 and 72 hours. MIT method was used to detect the effect of the formula on MC3T3-E1 multiplication activity.Besides,MC3T3-E1 were cultured respectively in 10% formula serum,pure serum and fetal bovine serum for 24,48 and 72 hours,and the flow cytometer was used to detect the effect of the formula on the MC3T3-E1 cycle. Results： compared to pure serum and fetal bovine serum,there was no significant difference between MC3T3-E1 multiplication activities and the cell cycles in the formula serum（ P 〉0. 05）. Conclusion： Kidney-tonifying Blood-activating and Tooth-astringing Formula has no effect on MC3T3-E1 multiplication activity and the cell cycle,but it may work by facilitating its osteoblast differentiation and promoting the alveolar bone regeneration.
