摘 要:目的研究红花抗早孕作用及对早孕小鼠子宫内膜血管内皮生长因子(VEGF)表达的影响,并观察其毒理作用。方法将60孕鼠平均分为红花低、中、高剂量组及空白对照组,每组15只。红花低、中、高剂量组分别按相当于生药量1、3、5 g/kg红花单品水煎剂灌胃,空白对照组每日予等容积蒸馏水灌胃。另选28只未孕雌鼠,分组及灌胃方法同孕鼠组。比较各孕鼠组止孕率及子宫内膜VEGF表达情况,并观察各未孕组小鼠体质量增长情况,评价红花毒理作用。结果红花低、中、高剂量组止孕率较空白对照组均明显升高,比较差异均有统计学意义(P<0.05),且随红花剂量增加止孕效果越明显(P<0.05);红花低、中、高剂量组孕鼠VEGF在子宫内膜IOD值均低于空白对照组,但仅红花中、高剂量组与空白对照组比较差异有统计学意义(P<0.05),红花低剂量组比较差异无统计学意义(P>0.05);红花低、中、高剂量组灌胃前后体质量增长率均低于空白对照组,但仅红花高剂量组与空白对照组比较差异有统计学意义(P<0.05),红花低、中剂量组比较差异无统计学意义(P>0.05)。结论红花能明显降低孕鼠子宫内膜VEGF的表达水平,影响胚胎血管的形成,从而起到抗早孕的作用,且无明显毒副作用。Objective To effect the anti - early pregnancy function of Honghua in early pregnant mice and its effect on vascular endothelial growth factor (VEGF) in endometrium, and to observe the toxicological effect. Methods 60 pregnant mice were averagely divided into Honghua low, middle, high dose group and blank control group, 15 mice in each group. Mice in Honghua low, middle, high dose group were received Honghua decoction which equivalent of 1,3, 5 g/kg of cure drug respectively, via gavage. Mice in blank control group were received equivalent volume distilled water via gavage. Then took another 28 unpregnant female mice which received same grouping method and gavage method as pregnant mice group. The terminating pregnancy condition and the expression of VFGF in endometrium were observed in each pregnant mice group. The increase of body mass was observed in unpregnant group, and the toxicological effect of Honghua was evaluated. Results The terminating pregnancy rates in Honghua low, middle and high dose group was obviously increased as compared with those in blank control group, there were significant differences ( P 〈 0.05 ), and the terminating pregnancy effect increase with dosage of Honghua (P 〈0.05). The integrated optical density (IOD) of endometrium VEGF in Honghua low, middle and high dose group was lower than that in blank control group, but the statistical difference on IOD only among Honghua middle, high dose group and blank control group ( P 〈 0.05 ) , there was no statistical difference on IOD between Honghua low dose group and blank control group ( P 〉 0.05 ). The growth rate of body mass before and after lavage in .Honghua low, middle and high dose group was lower than that in blank control group, but the statistical difference on bodymass only between Honghua high dose group and blank control group (P 〈 0.05 ), there was no statistical difference on on body mass among Honghua low, middle dose group and blank control group ( P 〉 0.05 ).
