肠道病毒71感染手足口病患者27种细胞因子的研究

摘　　要：目的基于液相芯片分析平台，揭示EV71感染手足口病患者细胞因子表达水平与病情严重程度的关系。方法收集济南市2009—2014年43例EV71感染手足口病患者血清，其中16例为轻症患者、27例为重症患者，同时随机选取10例健康人群血清作为对照组。利用Bioplex液相芯片平台检测分析血清中27种细胞因子表达水平。结果EV71感染手足口病患者重症组血清中22种细胞因子表达水平较对照组有不同程度升高（P〈0．02），包括IL-1ra、IL-2、IL-4、IL-5、IL-6、IL-7、IL-8、IL-9、IL-10、IL．12（p70）、IL-13、IL-15、IL-17、etotaxin、G—CSF、IFN-γ、IP-10、MCP-1、PDGF—BB、MIP-1β、RANTES和TNF-α；轻症组血清中21种细胞因子表达水平较对照组显著升高（P〈0．02）。GM-CSF在重症组和轻症组表达水平均较对照组明显降低（P〈0．02）。等级相关分析显示12种细胞因子表达水平与病情严重程度呈中度正相关（IL-1ra、IL-7、IL-9、IL-10、IL-13、IL-17、etotaxin、IP-10、PDGF—BB、MIP-1β、RANTES和TNF—α），GM—CSF与病情程度呈中度负相关。结论济南市EV71感染手足口病患者血清中多种细胞因子表达水平在病情发展过程中发生显著变化，并与病情严重程度密切相关。液相芯片为手足口病细胞因子研究提供了-种高效平台。Objective To detect and analyze the levels of 27 serum cytokines expression in EV71- induced Hand Foot and Mouth Disease（HFMD） in Jinan and revealed their correlations with disease severity based on the platform of liquid chip. Methods 43 serum samples were collected from EV71-infected HFMD patients in Jinan in 2009-2014,including 16 mild cases and 27 severe cases. 10 serum specimens from healthy people were also collected as controls. 27 serum cytokines were analyzed on Bio-Plex liquid chip platform. Results Compared to healthy controls, 22 cytokines expression levels increased in severe groups, including IL-1 ra, IL-2, IL-4, IL-5, IL-6, IL-7, IL-8, IL-9, IL-IO, IL-12 （ p70 ） , IL-13, IL-15, IL-17, etotaxin, G-CSF, IFN-γ, IP-IO, MCP-1, PDGF-BB, MIP-113, RANTES and TNF-α （ P 〈 0.02 ）. 21 eytokines expression levels elevated than healthy controls （ P 〈 0. 02）. GM-CSF was found decreased levels in both mild and severe groups than in the healthy controls （ P 〈 0. 02）. The correlation analysis showed that 12 eytokines （ IL-1 ra, IL-7, IL-9, IL-10, IL-13, IL-17, etotaxin, IP-10, PDGF-BB, MIP-1β, RANTES and TNF-α ） were moderately correlated with the severity of EV71-infected HFMD. GM-CSF was negatively correlated with HFMD. Conclusions Many kinds of serum cytokine changed obviously in the EV71-infeeted HFMD patients and cytokine levels were closely associated with the severity of HFMD. The analysis of liquid chip for serum cytokines provides an efficiently method.
