摘 要:目的从代谢组学角度探讨异常黑胆质成熟剂(ASMq)对抑郁症大鼠血浆代谢物的影响。方法选取60只雄性SD大鼠,进行21d慢性不可预见性温和应激(CUMS)诱导建立抑郁症大鼠模型(接受应激组),并通过体质量测量、糖水消耗实验、矿场实验进行宏观评价。造模成功后,将接受应激组大鼠随机分为5组:ASMq高、中、低剂量组(给药剂量分别为6.0、3.0、1.5 g/kg)及阳性对照组(氟西汀3.5 g/kg)、模型对照组(生理盐水1.5 mL/100g),每组12只。另设正常对照组(12只,不接受任何应激,正常饲养)。末次给药1h后颈椎脱臼处死大鼠,采用核磁共振技术检测6组大鼠血浆代谢物的变化。结果与抑郁模型对照组比较,ASMq低、中、高剂量组大鼠血浆中肉毒碱、乳酸、β-羟丁酸、肌酸、丙酮酸、丙氨酸、极低密度脂蛋白(VLDL)、络氨酸、糖蛋白、苯丙氨酸、丙酮、甲酸、不饱和脂类含量下降,差异有统计学意义(P〈0.05)。β-葡萄糖、组氨酸、苯丙氨酸、α-葡萄糖含量显著升高,差异有统计学意义(P〈0.05)。结论 ASMq可能通过对抑郁症大鼠的糖代谢、脂代谢、氨基酸代谢起到一定的调控和改善作用而提高机体兴奋性,治疗抑郁症。Objective To observe the effects of Abnormal Savda Munziq(ASMq) on endogenous metabolic changes in plasma in depression rat model to explore the internal mechanisms of ASMq in the point of view of metabonomic.Methods Depression model was established on 72 male SD rats using chronic unpredictable mild stress method(CUMs).Then,serum samples from all groups were analyzed by nuclear magnetic resonance technology.Results Compared to the control group,serum levels of isoleucine,lactic acid,creatine,glutamine,tyrosine,sugar,protein,fat,unsaturated lipids,β-hydroxybutyric acid,acetic acid,alanine,α-,β-glucose and glucose were significantly increased(P〈0.05) in the depression model group,while serum level of histidine was significantly decreased(P〈0.05).In contrast to the depression model group,serum level of histidine,α-glucose,β-glucose are significantly increased(P〈0.05) in the low,medium and high dose of ASMq groups,while serum levels of carnitine,lactic acid,β-hydroxybutyrate butyric acid,creatine,pyruvic acid,phenylalanine,alanine,lipid(VLDL,LDL),unsaturated lipids,acetone,tyrosine,glycoproteins and acid content were decreased significantly(P〈0.05) in all ASMq groups.Conclusion ASMq may improve the body excitability and treat depression in rats by improving and regulating the glucose,lipid and amino acid metabolism.
