摘  要:[目的]探讨翻白草丹参合剂对2型糖尿病(T2DM)大鼠模型过氧化物酶增殖受体γ(PPAR-γ)基因表达作用的分子机制.[方法]用小剂量链脲佐菌素(STZ)加高糖高脂饲料喂养大鼠制作T2DM大鼠模型.将大鼠随机分为正常对照组、模型对照组、阳性对照组和药物治疗组,分别灌胃给予蒸馏水、马来酸罗格列酮、翻白草丹参合剂12周.检测大鼠体质量、空腹血糖(FBC),总胆固醇(TG)、三酰甘油(TC)、高密度脂蛋白胆固醇(HDL-C)及低密度脂蛋白胆固醇(LDL-C)水平.并采用PT-PCR方法检测大鼠大网膜PPAR-γ基因表达.[结果]与正常对照组比较,模型对照组体质量显著降低(P<0.001),FBC、TC、TG、LDL-C值显著增高(P<0.001);与模型对照组比较,阳性对照组和药物治疗组体质量显著增高(P<0.05),FBC、TC、TG值显著降低(P<0.05),PPAR-γmRNA表达水平显著上调(P<0.05).[结论]翻白草丹参合剂可能通过提高PPAR-γ基因表达参与降低血糖及改善脂代谢紊乱.OBJECTIVE To study on molecular mechanism of the mixture of potentilla discolor bunge and salvia miltiorrhiza bunge on the expression of PPAR-γ gene in type 2 diabetic rat model.METHODS Type2 diabetic rat model was produced by high-fat diet with small dose of streptozotocin.Ninety rats were randomly divided into 4 groups of normal control,model,positive control and test,and were respectively performed by intragastric administration with water,rosiglitazone maleate,potentilla discolor bunge and salvia miltiorrhiza bunge for 12 weeks.The weight,fasting blood glucose(FBG),total cholesterol(TC),Triglyceride(TG),high density lipoprotein cholesterol(HDL-C)and low density lipoprotein cholesterol(LDL-C)were respectively detected,and the expression of PPAR-γ gene in the omentum majus was detected by RT-PCR.RESULTS The weight and the levels of FBC,TC,TG,LDL-C were significantly lower and higher in the model group than in the normal control group(P〈0.001),respectively,and as compared with model group,the weight and the levels of FBG,TC,TG were observably higher and lower in test group and positive control group(P〈0.05),respectively.The expression of PPAR-γ gene was lower in type 2 diabetic rat model group than in normal control group(P〈0.05),and higher in test group and positive control group than in negative group(P〈0.05).CONCLUSION The mixture of Potentilla discolor bunge,and salvia miltiorrhiza bunge maybe participate in reducing blood glucose and improving lipid metabolism disorders with increase of PPAR-γ gene expression.
