摘  要:[目的]探讨反复臭氧暴露对小鼠实验性变应性鼻炎的影响.[方法]将雌性BALB/c实验小鼠分为对照组、卵清蛋白喷雾组、卵清蛋白喷雾/臭氧暴露组、卵清蛋白全身致敏组及卵清蛋白全身致敏/臭氧暴露组,给予相应处理.用ELISA法测定总血清中IgE,anti-OVA IgE,anti-OVA IgG1含量及鼻腔灌洗液中IL-4,IL-5,INF-γ水平.用Luna染色法测定鼻黏膜中嗜酸性粒细胞浸润数量.[结果]单独卵清蛋白喷雾暴露诱导较弱过敏反应,与臭氧同时暴露可明显升高血清IgE,anti-OVA IgE,anti-OVA IgG1水平,从而加剧免疫反应,且显著增加鼻腔灌洗液中Th2细胞因子水平及鼻黏膜中嗜酸性粒细胞数目;卵清蛋白喷雾/臭氧暴露组血清IgE,anti-OVA IgE水平显著低于卵清蛋白全身致敏组(P<0.05);全身致敏小鼠卵清蛋白喷雾与臭氧同时暴露可加重过敏反应.[结论]臭氧暴露诱导或加重过敏反应,其可能机制是通过抗原特异性抗体及Th2细胞因子生成增多而引起.OBJECTIVE To study the effects of ozone exposure again and again on experimental allergic rhinitis in mice.METHODSFemale BALB/c mice were divided into control group,ovalbumin(OVA)-aerosol exposure group,OVA-aerosol/ozone exposure group,OVA systemic sensitization group,and OVA systemic sensitization/ozone exposure group,and were treated with the corresponding process.By using ELISA,it was measured that the concentration of total IgE,anti-OVA IgE,anti-OVA IgG1 in serum and IL-4,IL-5and INFγin nasal lavage fluid(NLF),and the number of eosinophils infiltrated in nasal mucosa were detected by the Luna staning.RESULTSOVAaerosol exposure alone induced weak allergic response in non-sensitized mice,and the combination of OVAaerosol and ozone exposure resulted in augmented immune responses to significantly increase of total IgE,anti-OVA IgE and anti-OVA IgG1 antibody titers,as well as significant increase of Th2 cytokines in NLF and the number of eosinophils infiltrated in nasal mucosa.The titers of total IgE and anti-OVA IgE were significantly lower in the OVA aerosol/ozone exposure group than in the OVA systemic sensitization group(P0.05),and the OVA aerosol/ozone exposure at the same time enhanced the allergic responses in systemic sensitized mice.CONCLUSION Ozone exposure induces and enhances the allergic responses by augmenting the production of allergen specific antibody and Th2 cytokines.
