摘 要:目的:观察急性冠脉综合征(ACS)患者血小板蛋白酶活化受体-1(PAR-1)的表达,探讨普伐他汀对血小板PAR-1表达的影响。方法:临床研究共纳入110例研究对象,分为急性冠脉综合征(ACS)组、稳定型心绞痛组和正常对照组,通过ELISA方法检测富血小板血浆PAR-1的表达。体外研究给予不同浓度普伐他汀干预及ADP刺激AMI患者和正常对照者血小板,采用流式细胞仪检测血小板PAR-1和LOX-1表达的变化。结果:ACS组PAR-1浓度显著高于稳定型心绞痛组和正常对照组;稳定性心绞痛组PAR-1浓度显著高于正常对照组。体外研究证实普伐他汀呈浓度依赖的方式抑制ADP诱导的血小板PAR-1和LOX-1表达。结论:ACS患者血小板表达PAR-1明显增加,普伐他汀体外干预能明显降低ADP诱导的进一步血小板PAR-1的表达。Objective To observe the expression of protease-activated receptor-1 (PAR-l) in patients with acute coronary syndrome (ACS) and to study the influence of pravastatin in vitro on PAR-1 expression. Methods A total of 110 patients were divided into ACS group, stable angina group and control group. The expression of PAR-1 in PRP was detected by ELISA. Different concentrations of pravastatin and ADP in vitro were for AMI patients and controls and flow cytometer was used for the expression change of PAR-1 and LOX-1. Results We had confirmed that PAR-1 expression in ACS group was significantly higher than that in stable angina group and control group. Treatment with pravastatin could inhibit the expression of PAR-1 and LOX-1 on platelets induced by ADP in a concentration dependent manner. Conclusions There is significant increase of PAR-1 expression in ACS patients and pravastatin could significantly inhibit the expression of PAR-1 and LOX-1 on platelets induced by ADP in a concentration dependent manner in vitro studies.
