摘 要:目的观察黄绿青霉素(citreoviridin,CIT)对血管内皮细胞(vascular endothelial cells,VEC)的毒性及硒(selenium,Se)的保护作用。方法体外培养人脐静脉血管内皮细胞(human vascular endothelial cell,HUVEC),以亚硒酸钠(Na_2SeO_2)和黄绿青毒素(CIT)处理,分为:对照;Se(培养基含Se元素0.1mg/L);CIT低剂量(含0.2mg/L CIT);CIT高剂量(含0.4 mg/L CIT);Se+CIT(0.2mg/L Se和0.4mg/L CIT)5组。药物作用12 h后分别用MTT法检测细胞增殖活性,AnnexinV-FITC/PI双染法检测细胞凋亡,流式细胞仪检测细胞周期。结果 CIT组细胞增殖活性低于对照组,Se+CIT组细胞增殖活性高于CIT组(P<0.05);CIT组细胞凋亡率高于对照组,Se+CIT组细胞凋亡率低于CIT组(P<0.05);CIT组细胞发生G0/G1期细胞周期阻滞,Se+CIT组G0/G1期细胞周期阻滞不明显。结论 CIT可促进细胞凋亡,通过G0/G1期细胞周期的阻滞作用抑制HUVECs增殖活。Objective To investigate the effects of citreoviridin (CIT) on cell proliferation and apoptosis in human umbilical vascular endothelial cells (HUVECs), and the potential protective effect of selenium (Se). Methods HUVECs were cultured in DMEM medium, and the cells were divided into 5 groups: control: Se model (0.1 mg/L Se): low dose CIT model (0.2 mg/L CIT): high dose CIT model (0.4 mg/L CIT): Se + CIT model (0.1 mg/L Se and 0.4 mg/L CIT). Cell proliferation viability was tested using MTT assay. Annexin V-FITC/PI staining method was used to detect the level of cell apoptosis. The effects of CIT and Se on cell cycle were determined using FCM method. Results The levels of cell viability in CIT groups were lower than the control, while the level in Se+CIT group was higher than CIT groups (P〈0.05) .The G0/G1 phase accumulation of cell cycle was observed in both low dose CIT group and high dose CIT group, while the effect of G0/G1 phase accumulation was not different significantly in Se+CIT group.The levels of cell apoptosis in CIT groups were higher than in control group, while the level in Se+CIT group was lower than the CIT groups (P〈0.05) . Conclusion CIT induced cell apoptosis in HUVECs and inhibited cell proliferation via the G0/G1 phase accumulation of cell cycle. The toxic effects of CIT on HUVECs were restrained by the treatment of Se.
