摘 要:目的初步探讨1,25-二羟基维生素D3(1,25。(0H)zD3)对链脲佐菌素(STZ)所致大鼠胰岛素瘤(INS-1)细胞损伤的保护与修复作用。方法将INS-1细胞接种于培养板中,置于CO:培养箱中进行培养,药物干预后,应用细胞计数试剂盒(CCK8)检测细胞增殖活力,ELISA法分别检测培养液中的胰岛素浓度、丙二醛(MDA)浓度和总抗氧化能力(T-AOC)。结果1,25-(0H):D3可以促进INS-1细胞的活力和胰岛素的分泌(P〈0.01)。STZ所致模型组的细胞活力降低(P〈0.001),胰岛素分泌量减少(P〈0.01),MDA含量明显升高(P〈0.01),T-AOC能力下降(P〈0.01)。与STZ组比较,1,25-(OH)2D3保护组与修复组在不同程度上改善了上述指标(P〈0.01)。结论1,25-(oH)2D3可以促进INS-1细胞分泌胰岛素,对STZ损伤的胰岛细胞具有-定的保护与修复作用。Objective To explore the protection and repair effects of 1,25-dihydroxyvitamin D3 on STZ-induced INS-1 cells damage. Methods After inoculating the INS-1 cells into the culture plates, the proliferation activity of the cells was measured by CCK8 method. Secreted insulin and malondialdehyde (MDA) concentration, total anti-oxidation capability (T-AOC) in the culture medium were measured by ELISA. Results 1,25-dihydroxyvitamin D3 improved the viability and insulin secretion of the INS- 1 cells(P〈0.01). The cell proliferation (P〈0.001), insulin secretion (P〈0.01) and T-AOC (P〈0.01) of the STZ group decreased, and the MDA content (P〈0.01) significantly increased Compared with the STZ group, the protection and repairation group of 1,25-dihydroxyvitamin D3 can improve the above index in different degrees(P〈0.01). Conclusions 1,25-dihydroxy vitamin D3 improved the insulin secretion of the INS-1 cells and had significantly protection and repair effects on the INS-1 cells damaged by STZ.
