摘 要:目的探讨兴海县鼠疫生物学特性及流行病学意义,为鼠疫防治提供科学依据。方法对1956-2009年兴海县自动物及人间分离的鼠疫菌进行糖(醇)类酵解、毒力测定、质粒检查、基因型等实验,对该疫源地分离的鼠疫菌株进行分析。结果对15株鼠疫菌的糖(醇)类酵解试验观察,除自五趾跳鼠分离的菌株鼠李糖为阳性外,其余14株均为青藏高原型;被试菌株均能产生鼠疫菌荚膜抗原(F1)及鼠疫杆菌素I(Pst I);毒力抗原因子(VW)阳性的菌株占80.00%(12/15);色素沉着因子(Pgm)阳性的菌株占93.33%(14/15);质粒检查除五趾跳鼠分离的菌株为6×10^6、29.19×10^6、65×10^6,1962年人尸分离的鼠疫菌带有6×10^6、27.04×10^6、65×10^6质粒外,其余菌株均为6×10^6,45×10^6,52×10^6;基因型除五趾跳鼠和1962年人尸上分离的菌株为21型外,其余均为8型;毒力检测结果显示所测菌株80.00%为强毒株。结论兴海县喜马拉雅旱獭鼠疫疫源地仍处于动物鼠疫流行的活跃期,应加强鼠疫监测和预警,控制人间鼠疫的发生。Objective To investigate the biological characteristics and epidemiological significance of Yersinia pestis strains from Xinghai county in Qinghai Province in order to provide a scientific basis for plague prevention and contro1. Methods To analyze the strains about glycolysis testing,virulence,plasmid,genotype and other experiments were determined on Yersinia pestis isolated from 1956 to 2009 of animal and human in Xinghai county of Qinghai Province. Results According to glycolysis testing of 15 Yersinia pestis strains,in addition to one strain separated from Allactage sibirica rhamnose was positive,the remaining 14 strains were Qinghai-Tibet plateau type; all the strains could produce F1 and Pst1; the virulence factor VW positive strains accounted for 80%( 12 /15); Pgm positive strains accounted for 93. 33%( 14 /15); plasmid test showed that in addition to the strains separated from Allactage sibirica was 6 × 10^6,29. 19 × 10^6,65 × 10^6,strains from dead people in 1962 was 6 × 10^6,27. 04 × 10^6,65 × 10^6,the rest of the strains were 6 × 10^6,45 ×10^6,52 × 10^6; the genotype in addition to the strains separated from Allactage sibirica and dead people in1962 were genomovar 21, the rest were genomovar 8; toxity test showed 80% strains were velogenic. Conclusions Himalayan marmot animal plague foci in Xinghai county is still active,should strengthen monitoring and early warning to control the occurrence of human plague.
