摘  要:使用不同浓度的NaCl溶液处理野大麦,利用透射电镜观察野大麦叶肉细胞超微结构对不同浓度NaCl胁迫的响应。结果表明:对照组的叶肉细胞排列疏松,各种细胞器具有完整的结构,少量淀粉粒和脂质球存在于叶绿体中。低浓度NaCl(150mmol/L、350mmol/L)对叶肉细胞超微结构具有较小的影响。中等浓度NaCl(550mmol/L)和高浓度NaCl(750mmol/L)使叶肉细胞超微结构发生变化,叶绿体中的类囊体肿胀,叶绿体的形状由原来的梭形或长椭圆形变成圆球状,片层结构逐渐消失,脂质球增多,叶绿体内、外膜逐渐消失直至叶肉细胞中看不到叶绿体的存在;部分线粒体中的嵴消失,基粒紊乱;野大麦叶肉细胞细胞膜系统结构逐渐消失解体。The ultra structural changes of mesophyll cells in Hordeum brevisubulatum under NaCl stress with different concentration were observed using transmission electron microscopy.The results showed that the leaf mesophyll cells in control group of Hordeum brevisubulatumarranged loosely,various organelles were morphologically integrated,some starch grains and electron-dense globules were presented in chloroplasts.No alteration of ultrastructure was found under NaCl stress with low concentration(150mmol/L、350mmol/L).While under NaCl stress with medium concentration(550mmol/L)and high concentration(750mmol/L),the most conspicuous change was the swelling of the thylakoid membranes,the shape of the chloroplast changed from fusiform or oblong to spheroid,the lamellar structure of the chloroplast were disorganized gradually,the number of plastoglobulus increased obviously,the inner and outer membrane of the chloroplast disappeared gradually until the chloroplast disappeared completely;the breakdown of cristae were observed in some mitochondria,disorganization of grana;the membrane system structure of Hordeum brevisubulatum mesophyll cell disappeared gradually.
